摘要
【目的】确定从酸枣仁中提取总生物碱的最适方法和最佳提取条件。【方法】在单因素优化试验的基础上采用正交试验设计,比较优化了溶剂回流法提取酸枣仁总生物碱的工艺条件。【结果】溶剂回流法提取酸枣仁总生物碱的最佳条件为:以乙醇为提取溶剂,乙酸辅助提取,质量分数为20%乙酸的乙醇溶液(即,乙醇∶乙酸=80∶20(v/v)),以20∶1的液固比,在60℃下回流提取4.0h。【结论】该方法得到的提取物较之正交优化前酸枣仁提取物中的总生物碱含量可提高60%以上,且获得的总生物碱粗提物得率可达11%。
【Objective】To obtain the optimum extraction conditions of total alkaloids from Zizyphi Spinosi Semen(ZSS),【Methods】single factor experiment and orthogonal experimental design were applied,respectively.【Results】The results were as follows:20% acetic acid resolved in ethanol as extracting solvent(EtOH∶CH3COOH = 80∶20(v/v)),ratio of liquid to solid 20∶1(mL/g),reflux for 4.0hrs at 70 ℃.【Conclusion】The contents of total alkaloids in extract from ZSS with the optimum extraction conditions above were over 60% more than the extract without the optimization.And recoveries of total alkaloids extract from ZSS arrived 11%.This method provided a reference for the studying ingredients and activities of alkaloids in ZSS.
