摘要 [目的]谷子是抗旱研究的模式植物,谷子基因组测序的完成,为分离鉴定谷子抗旱基因提供了便利,WRKY基因参与植物非生物胁迫应答,在植物抗旱方面也发挥重要作用。为分离鉴定谷子WRKY类抗旱转录因子基因,从而为谷子抗旱机理研究和抗旱育种提供参考。[方法]采用生物信息学方法,对谷子转录因子SiWRKY基因家族进行了分离鉴定和表达分析。[结果]我们发现,谷子基因组中存在103个WRKY基因,分布在谷子的9条染色体上,根据在染色体上的位置命名为SiWRKY1-SiWRKY103。[结论]80%的谷子WRKY基因在干旱胁迫后上调表达,表明其参与谷子抗旱机制,为植物抗旱研究提供了候选基因。 [Objective]WRKY transcription factor is one of the biggest gene families that are important regulators of major plant processes,including growth,development,and responses to stress.However,there is limited information about the numbers,structures and functions of those WRKY transcription factors in foxtail millet(Setaria italica(L.)P.Beauv.).[Methods]In this paper,the WRKY transcription factor gene family in foxtail millet were separated and identified by bioinformatics,and the expression were analyzed by qRT-PCR.[Results]In our study,103 WRKY transcription factor-encoding genes were identified in the S.italica genome.The genes were named SiWRKY1 to SiWRKY103 according to their order on the chromosomes.The expression among four different tissues was analyzed using publicly available RNA sequencing data.The expression of SiWRKYgenes in response to dehydration were analyzed by qRT-PCR.[Conclusion]In conclusion,most SiWRKYgenes(〉80%)were up-regulated by drought stress,it may indicated that these genes play very important roles for drought-resistant in millet,and our conclusion will provide a set of candidates genes for the research of plants' response to drought stress.
