摘要
[目的]探析谷子调控干旱胁迫的E3基因。[方法]本文基于二代测序技术获得勾勾母鸡咀(GG)和晋汾16(JF)在PEG胁迫下的转录表达谱数据,运用生物信息学检索筛选出11个干旱相关的E3基因,对其理化性质、功能域、启动子元件、表达模式以及与其它物种E3基因的系统进化分析。[结果]蛋白等电点介于4.78~8.88之间,功能域主要为ZF_UBR型、ZF_RING和U_BOX型三类,启动子序列包含干旱诱导响应元件MBS或脱落酸响应元件ABRE等,其中Si001665m和Si022170m与PUB22/23亲缘关系较近,Si029090m与PUB19亲缘关系较近。[结论]初步确定这些是调控谷子干旱胁迫的E3候选基因,可能受干旱或脱落酸诱导表达,这为进一步研究谷子干旱响应相关的泛素连接酶E3基因提供理论依据。
[Objective]Analysis of E3 genes in foxtail millet under drought stress.[Methods]In this paper,Gou Gou Mu Ji Ju and Jin Fen 16 based on expression profiles under PEG stress using NGS technology,11 differentially expressed ubiquitin ligase E3 genes were revealed.Through the prediction and analysis of physicochemical properties,functional domains,regulatory elements in promoters,expression difference,sequence alignment and phylogenetic analysis.[Results]We found that isoelectric point was between 4.78~8.88,functional domains mainly include ZF_UBR type,ZF_RING type,and U_BOX type.The promoters sequence contained MBS,ABRE,etc.Phylogenetic showed that Si001665 m and Si022170 m were closely related to PUB22/23,Si029090 m were closely ralated to PUB19.[Conclusion]Some ubiquitin ligase E3 genes,which may be induced by drought or ABA,were identified.This would provide a theoretical basis for further research of ubiquitin ligase E3 in response to drought in foxtail millet.
