半乳糖凝集素-3和基质金属蛋白酶-9在宫颈癌组织中的表达及其临床意义

Expression and Significance of Galectin-3 and MMP-9 protein in cervical carcinoma

目的探讨半乳糖凝集素-3(Galectin-3)和基质金属蛋白酶-9(MMP-9)在宫颈癌组织中的表达及临床意义。方法用免疫组化SP法检测15例正常宫颈组织、15例宫颈上皮内瘤变组织、48例宫颈癌组织中Galectin-3和MMP-9蛋白的表达情况,并结合临床病理因素进行分析。结果 Galectin-3蛋白在宫颈癌组织中的阳性表达率明显高于正常宫颈组织及宫颈上皮内瘤变组织,MMP-9蛋白在宫颈癌组织中的阳性表达率也明显高于正常宫颈组织和宫颈上皮内瘤变组织,差异均有统计学意义(P<0.01)。在宫颈癌组织中,Galectin-3蛋白阳性表达率随病理分级的升高而逐渐升高,有淋巴结转移者高于无淋巴结转移者;同时MMP-9蛋白阳性表达率随病理分级的升高而升高,有淋巴结转移者大于无淋巴结转移者,差异均有统计学意义(P<0.05)。在宫颈癌组织中,Galectin-3蛋白和MMP-9蛋白表达呈正相关(P<0.01)。结论应用免疫组化的方法检测Galectin-3和MMP-9在宫颈癌中的表达及其临床意义、相关性研究,探讨其在宫颈癌发生发展中的意义,并对宫颈癌治疗提供新的思路。MMP-9蛋白和Galectin-3蛋白的异常表达对宫颈癌细胞恶性转化、增殖和浸润转移有一定的生物学意义,它们之间的这种正反馈效应可能在宫颈癌的发生发展过程中发挥重要的作用,至于其在宫颈癌中发生发展过程中的作用机制需今后进一步研究。

Objective To study the expression and significance of Galectin-3 protein and MMP-9 protein in cervical carcinoma. Methods Immunohistochemical sp staining method was used to detect the expression of Galectin-3 protein and MMP-9 protein in 15 cases of normal cervical epithelium, 15 cases of cervical intraepithelial neoplasia, and 48 cases of cervical carcinoma. The results were analyzed combining with clinicopathological data. Results The expression of Galectin-3 protein was apparently higher in cervical carcinoma than in normal cervical epithelium and cervical intraepithelial neoplasia, the expression of MMP-9 protein was also apparently higher in cervical carcinoma than in normal cervical epithelium and cervical intraepithelial neoplasia, the difference was statistically significant （P 〈 0.01 ）. In cervical carcinoma tissues, the positive expression rate of Galectin-3 protein elevated along with pathologic stage, and the rate in patients with lymphatic metastasis is higher than that in patients without lymph node metastasis, the same tendency was observed for MMP-9 protein ,the difference was statistically significant（ P 〈 0.05 ）. The positive correlation was also found between Galectin-3 and MMP-9 protein expression in cervical carcinoma（ P 〈 0.01 ）. Conclusion The abnormal expression of Galectin-3 and MMP-9 is involved in the cervical carcinoma cells proliferation, malignant transformation,invasion and metastasis. They may play an important role during the occurrence and development of human cervical carcinoma, and the mechanisms need to be further studied.
