红豆杉多糖对免疫低下小鼠血清IL-2、IFN-γ的影响 被引量:7

Effects of Taxus polysaccharide on serum IL-2 and IFN-γ of immunosuppression mice

摘要 目的探讨红豆杉多糖调节免疫低下小鼠免疫功能的机制。方法将48只小鼠采用随机数字表法分成为空白组、模型组及红豆杉多糖低、中、高剂量组和阳性对照组,每组8只。除正常对照组外,其余组小鼠皮下注射氢化可的松0.2 mg/kg,隔天1次,共14 d,制备免疫低下模型。空白组和模型组灌胃0.9%Na Cl溶液200 mg/kg,红豆杉多糖低、中、高剂量组分别按红豆杉多糖100、200、400 mg/kg,阳性组按猴头菇多糖11 mg/kg灌胃连续14d。采用酶联免疫吸附法检测小鼠血清中白细胞介素-2(interleukin-2,IL-2)、干扰素-γ(interferon-gamma,IFN-γ)的含量;流式细胞术检测小鼠血CD4^+、CD8^+T细胞数量。结果 1模型组与空白组比较,血清中IL-2、IFN-γ含量均明显降低(P<0.01)。红豆杉多糖各剂量组及阳性组与模型组比较,血清中IL-2含量均有所增加(P<0.05),其中高剂量组明显增高(P<0.01);红豆杉多糖各剂量组、阳性组与模型组比较,红豆杉多糖高、中剂量组及阳性组血清中IFN-γ均明显增加(P<0.01);2模型组与空白组对比CD4^+、CD8^+T淋巴细胞均明显降低(P<0.01),红豆杉多糖各剂量组与模型组对比,CD4^+、CD8^+T淋巴细胞均有所增加(P<0.05)。结论红豆杉多糖调节免疫功能机制可能通过调节体液免疫来增强免疫低下小鼠的免疫功能。 Objective To explore the mechanism of the Taxus polysaccharide regulating immune function of immunosuppression mice. Methods Forty-eight male mice were randomly dividend into control group, model group, taxus polysaccharise group( low,medium and high does group) and positive control group. Mice in the control group were injected with 0.9% NaCl solution, other were injected with 0.2 mg/kg hydroeortisone every day for 14 days to establish the immuno- compromised model. The control group and model group were with 200 mg/kg normal saline, three Taxus polysaccharide groups were treated with Taxus polysaccharide( 100,200 and 400 mg/kg by body weight) ,one positive control group was treated with polysaccharide of hericium erinaceus. The cellular IL-2 and IFN-γ of mice was tested by Enzyme-linked immunoassay detection, The number of serum CD4+ and CD8+ were detected by flow cytometry. Results (2)The serum of IL-2,IFN-γ in model group was significant lower as compared with control group (P 〈 0.01 ), Compared with model group, the levels of IL-2 in the treatment groups were increased (P 〈 0.01 ). Compared with model group, the serum of IFN-γ of the high, middle close group had increased, with obvious significance in addition to the low dose group;(2)The number of CD4 + T cells and CD8 + T cells in model group were obviously reduced as compared with control group ( P 〈 0.05 ) ; The number of CD4+ T cells and CD8 + T cells in high-dose and middle-dose groups had obviously increased as compared with model group ( P 〈 0.01 ). Conclusion The mechanism of Taxus polysaccharide regulating the immune function in immunodepressed mice may associate with improvement immune function through regulating humoral immunity.
