摘要
实验以斑马鱼胚胎为研究对象,考察不同浓度的4-氯苯酚(1 mg/L、5 mg/L、10 mg/L)对斑马鱼胚胎的急性毒性及细胞凋亡.分别在24 h、48 h、72 h、96 h、120 h时测定胚胎的急性发育指标及细胞凋亡.结果显示,在不同浓度的4-氯苯酚溶液中,斑马鱼的孵化率随着暴露剂量的增加而降低,斑马鱼的死亡率与畸形率随着暴露剂量的增加而升高,形态学异常的主要表现为心包囊肿、脊柱弯曲、卵黄囊畸形等多种毒性效应;同时AO染色法以及细胞凋亡酶的结果显示4-氯苯酚能够诱导胚胎产生细胞凋亡.
The study was to investigate the acute toxicity of 4-chlorophenol to zebrafish embryos. The indicators included developmental toxicology and apoptosis were determined at 24 h,48 h,72 h,96 h and 120 h. The results showed as followed: accompanition with the increasing exposures concentrations,the survival rates and hatching rates were decreased,the malformation rates were increased and the hearting rates were accelerated.Moreover,the 4-chlorophenol exposures induced kinds of morphologic abnormalities,such as pericardium edema,spinal curvature and yolk sac edema. At the same time,the results of acridine orange stain and the activity of caspase-9 suggested 4-chlorophenol induced apoptosis to zebrafish embryos.
