摘要
目的探讨凝血酶抑制剂nexin-1对脑出血大鼠脑神经损伤和血脑屏障通透性的影响。方法将大鼠随机分为对照组、模型组和干预组,采用尾状核注射自体动脉血的方法复制脑出血模型,在造模的过程中往干预组尾状核部位注射凝血酶抑制剂nexin-1,其余组注射生理盐水。在给药12h、24h和36h后分别进行脑组织形态学检查、脑组织神经细胞凋亡检查、脑组织含水量检查和脑组织血脑屏障通透性检查。结果干预组脑神经细胞排列有轻微紊乱,形态结构异常的程度较轻,细胞核仍然有胞核固缩和胞质凝集的现象,但程度与模型组相比显著降低;干预组脑组织凋亡神经细胞数显著低于模型组(P<0.05);干预组脑组织含水量显著低于模型组(P<0.05);干预组大鼠脑组织皮层和基底节的血脑屏障通透性显著低于模型组(P<0.05)。结论凝血酶抑制剂nexin-1对脑出血后的脑神经损伤、血脑屏障通透性起保护作用。
Objective To explore the effects of thrombin inhibitor nexin-1 on brain nerve damage and blood-brain barrier (BBB) permeability in rats with cerebral hemorrhage. Methods The model of intracerebral hemorrhage (ICH) was duplicated and established by infusing autologous arterial blood into caudate nucleus of rats which were randomly divided into control group,model group and intervention group. In the process of molding, the intervention group also received injection of thrombin inhibitor nexin-1, however other two groups received normal saline. Then 12 hours, 24 hours and 36 hours after administration, we respectively detected morphological examination,neuronal apoptosis, water content and BBB permeability of brain tissues. Results The intervention group showed slighter disorder in neuron arrangement, milder abnormal morphology and lower degree of nucleus condensation and cytoplasm aggregation compared with the other two groups. And the number of neuronal apoptosis, water content and BBB permeability of brain tissues in the intervention group were significantly lower than those in the model group (P〈0.05). Conclusion Thrombin inhibitor nexin-1 has protective effects on brain nerve damage and blood-brain barrier (BBB)permeability after cerebral hemorrhage.
