调控CD147基因表达的MicroRNA分子鉴定及其抗膀胱癌转移研究 被引量:1

摘要 目的 探讨调控CD147基因表达的Micro RNA对膀胱癌的增殖、迁移和浸润的关联性。方法 选取自2015年1月~2016年5月在我院接受治疗的膀胱癌患者50例,分别取膀胱癌组织以及癌旁组织采用免疫组化进行CD147阳性检测,构建CD147 si RNA重组腺病毒(Ad-CD147 si RNA)载体转染膀胱癌细胞,将CD147阳性表达的膀胱癌组织随机分为Ad-CD147 si RNA转染的干扰组,腺病毒颗粒转染的阴性对照以及未加处理的对照组,采用MTT检测肿瘤细胞增殖能力、采用划痕实验检测肿瘤细胞迁移能力、采用明胶酶谱实验检测MMP-2和MMP-9含量、采用Transwell小室法检测肿瘤细胞浸润能力。结果 CD147在膀胱癌组中阳性率90.00%(45/50)显著高于癌旁组织12.00%(6/50),差异有统计学意义(P〈0.05)。不同性别和不同年龄间CD147阳性表达比较,差异无统计学意义(P〉0.05);而不同的TNM分期及肿瘤细胞分化程度与CD147阳性表达比较,差异有统计学意义(P〈0.05)。空白组及对照组的膀胱癌细胞迁移距离、侵入基底胶数量、MMP-2以及MMP-9含量明显高于干扰组,体外膀胱癌细胞在490 nm处吸收值显著低于干扰组,差异有统计学意义(P〈0.05)。结论 CD147基因所表达的Micro RNA能够通过促进MMP-2和MMP-9的分泌而加速膀胱癌组织的血供形成,加速增殖、迁移以及浸润。 Objective To investigate the correlation of MicroRNA that regulates the expression of CD147 gene with the proliferation, migration and infiltration of bladder cancer. Methods A total of 50 patients with bladder cancer who were treated in our hospital from January, 2015 to May, 2016 were selected, and immunohistochemistry was used to detect the positive expression of CD147 in bladder cancer tissues and adjacent tissues, and vector-transfected bladder cancer cells of CD147 siRNA recombinant adenovirus(Ad-CD147 siRNA) were constructed. CD147 positive bladder cancer tissues were randomly divided into interference group transfected with Ad-CD147 siRNA, negative control group trans- fected by adenovirus particles, and untreated control group. MTI" was used to detect the proliferation of tumor ceils, the migration ability of tumor cells was detected by scratch test, the contents of MMP-2 and MMP-9 were detected by gelatin zymography, and tumor cell infiltration ability was measured by Transwell chamber method. Results The positive rate of CD147 in bladder cancer group was 90.00% (45/50), which was significantly higher than that in para-cancerous tissue(12.00%, 6/50). The difference was statistically significant(P〈0.05). There was no significant difference of CD147 expression between different genders and ages(P〉0.05). The expression of CD147 was significantly different between T stage and tumor cell differentiation(P〈0.05). The migration distance, invasion matrix quantity, MMP-2 and MMP-9 of bladder cancer cells in blank group and control group were significantly higher than those in the interference group, and the absorbance of bladder cancer cells at 490 nm was significantly lower than that in the interference group, the differences were statistically significant(P〈0105). Conclusion MicroRNA expressed by CD147 gene can accelerate the formation of blood supply, accelerate proliferation, migration and infiltration of bladder cancer by promoting the secre- tion of MMP-2 and MMP-9.
