摘要
目的探讨免疫上游因子TLR9、MyD88、NF-κB及免疫下游细胞因子IL-6、TNF-α在HSP组与正常对照组间的差异及相关性。方法采用实时荧光定量PCR检测HSP患儿外周血单核细胞TLR9、MyD88及NF-κB mRNA表达量,ELISA法检测血浆IL-6、TNF-α水平。结果 1HSP患儿外周血单核细胞TLR9、MyD88及NF-κB mRNA表达均显著高于正常对照组;2HSP组血浆IL-6、TNF-α水平较对照组水平显著增高;3HSP组中TLR9与MyD88、TLR9与NF-κB、MyD88与NF-κB、NF-κB与IL-6以及NF-κB与TNF-α均存在线性正相关关系。结论HSP患儿TLR9、MyD88、NF-κB、IL-6、TNF-α水平均较正常儿童显著升高。TLR9-MyD88-NF-κB信号通路参与HSP发病,并在其中起重要作用。
Objective To investigate the differences and correlations of immunological upstream factors of TLR9, MyD88, NF-κB and immunological downstream factors of IL-6 and TNF-α in the HSP group and normal control group. Methods The expression of TLR9, MyD88 and NF-KB in peripheral blood mononuclear cells(PBMC) of HSP were detected by real-time PCR. The levels of IL-6 and TNF-α were measured by ELISA. Results ①The expression of TLR9, MyD88 and NF-KB mRNA in peripheral blood mononuclear cells of children with HSP was significantly high- er than that of normal controls; ②The levels of IL-6 and TNF-α in plasma of HSP group were significantly higher than those in the control group; ③There was a positive linear correlation between TLR9 and MyD88, TLR9 and NF-κB, MyD88 and NF-KB, NF-KB and IL-6, NF-κB and TNF-α in HSP group. Conclusion The levels of TLR9, MyD88, NF-κB; IL'6 and TNF-α in children with HSP are significantly higher than those in normal children. TLR9-MyD88- NF-κB signal pathways play an important role in the pathogenesis of HSP.
