摘要
在家畜病理学教学中和一些病理诊断中,常规H.E.染色切片质量的好坏直接影响病理评判结果。一些切片由于保存时间长久会逐渐褪色,使得显微镜下结构模糊、失真,影响病理疾病诊断的准确性。为了有效利用陈旧褪色病理H.E.染色组织切片,探讨陈旧褪色病理H.E.染色组织切片重新利用技术,采用4种方法(酸性高锰酸钾氧化-草酸漂白法、盐酸乙醇脱色法、氨水脱色法和常规H.E.染色法)对塔里木大学动物科学学院基础兽医实验室陈旧褪色的病理组织切片进行复染。结果表明:酸性高锰酸钾氧化-草酸漂白法复染的组织切片显微镜下细胞核呈蓝紫色,细胞质呈粉红色,复染效果最好;复染过程中酸性高锰酸钾氧化-草酸漂白法的掉片数最少,显著低于其他3种方法(P<0.05);4种复染方法间组织完整率差异不显著(P>0.05)。说明4种复染方法中酸性高锰酸钾氧化-草酸漂白法的复染效果最好。
Whether the quality of conventional HE-stained pathological sections is good or bad,it directly affects the results of pathological diagnosis in livestock pathology teaching and some pathological diagnoses. Some sections will gradually fade due to long time to save,causing the indistinct structure and distortion under a microscope,thereby affecting the accuracy of pathological diagnosis of diseases. To effectively utilize the old and faded HE-stained pathological tissue sections and investigate the reuse technology of the old and faded HE-stained pathological tissue sections,four methods including acidic potassium permanganate oxidation and oxalic-acid bleaching,hydrochloric acid and ethanol decoloration,ammonia water decoloration,and conventional HE staining were used for the counterstaining of the old and faded HE-stained pathological tissue sections in the basic veterinary laboratory of the college of animal science of Tarim University. The results showed that acidic potassium permanganate oxidation and oxalic-acid bleaching had the best counterstaining effect on the counterstained tissue sections,showing blue-purple nuclei and pink cytoplasm under the microscope; the number of dropped sections using acidic potassium permanganate oxidation and oxalic-acid bleaching was minimum during the process of counterstaining,which was significantly lower than( P < 0. 05) that using other three methods; there were no significant differences( P > 0. 05) in the integrity rate of tissues among four counterstaining methods. The results indicate that the counterstaining effect of acidic potassium permanganate and oxalic-acid bleaching was best( P < 0. 05) among the four counterstaining methods.
