摘要
Clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated(Cas)9系统由于其突变效率高、制作简单、成本低,被越来越广泛地应用于植物基因组的精确修饰,是一种具有广阔应用前景的基因组定点改造分子工具。本研究构建了玉米淀粉合成酶调控因子基因ZmRSR1的CRISPR/Cas9载体系统,为在玉米中实现该基因的定向突变奠定了坚实基础,同时也为玉米基因组的定点突变修饰提供了重要参考。
As its characteristics of high efficiency,easy manipulation and low cost,the clustered regularly interspaced short palindromic repeats( CRISPR)/CRISPR- associated( Cas) 9 system has been widely used in precise modification of plant genome,which is regarded as a molecular tool with a broad application prospect. A detailed protocol of targeted mutagenesis of starch synthase regulatory factor gene ZmRSR1 in maize using the CRISPR/Cas9 system was provided,which might give a reference for site- directed modification of maize genome.
