幽门螺旋杆菌和p14^(ARF)在蒙古族、汉族胃癌患者癌组织中的表达变化及意义 被引量:1

摘要 目的探讨幽门螺旋杆菌(Hp)和抑癌基因p14^(ARF)在蒙古族、汉族胃癌患者癌组织、癌旁组织和正常组织中的表达变化及意义。方法采用免疫组化SP法、改良的Warthin-Starry(W-S)银染色法和逆转录聚合酶链反应(RT-PCR)法分别检测Hp和p14^(ARF)在27例蒙古族胃癌患者和30例汉族胃癌患者癌组织、癌旁组织和正常组织中蛋白和mRNA表达。Spearman相关性分析汉族、蒙古族胃癌患者癌组织中Hp与p14^(ARF)表达的关系。结果p14^(ARF)在蒙古族、汉族胃癌患者癌组织中的表达低于癌旁组织和正常组织,差异有统计学意义(P均<0.05),癌旁组织低于正常组织,差异有统计学意义(P<0.05),p14^(ARF)在蒙古族、汉族人群各组织中的表达差异无统计学意义(P均>0.05)。蒙古族、汉族胃癌患者癌组织中Hp阳性率较高,两民族感染率比较差异无统计学意义(P>0.05)。汉族胃癌患者p14^(ARF)mRNA相对表达量在癌、癌旁、正常组织中两两比较差异均有统计学意义(P均<0.05)。蒙古族胃癌患者p14^(ARF)mRNA相对表达量在癌、癌旁、正常组织中两两比较差异均有统计学意义(P均<0.05)。蒙古族、汉族胃癌患者p14^(ARF)mRNA相对表达量在癌、癌旁和正常组织中差异无统计学意义(P均>0.05)。Spearman相关性分析显示,汉族、蒙古族胃癌患者癌组织中Hp与p14^(ARF)阳性表达呈负相关(r分别为-0.406、-0.433,P均<0.05)。结论蒙古族、汉族胃癌患者癌组织中p14^(ARF)均呈低表达,Hp阳性率均较高,Hp阳性与p14^(ARF)表达呈负相关关系;Hp与p14^(ARF)在蒙古族、汉族胃癌中的发病机制可能相同,二者可能共同促进胃癌的发生。 Objective To detect the expression of Helicobacter pylori (Hp)and p14ARF in the gastric cancer tissues, paracancerous tissue and normal tissues of patients with gastric carcinoma in Han and Mongolian nationalities and the mech-anism of two factors in the occurrence of gastric cancer.Methods Immunohistochemical (S-P)method,a modified War-thin-Starry (W-S)silver staining method and reverse transcription polymerase chain reaction (RT-PCR)were used to de-tect the expression of protein and mRNA of Hp and p14ARF in gastric cancer tissues,paracancerous tissues and normal tis-sues from 27 cases of Mongolian patients and 30 cases of Han patients with gastric cancer.Spearman analysis was used to analyze the correlation between the Hp and p14ARF expression in the gastric cancer tissues of patients from two nationalities. Results The expression of p14ARF in gastric cancer tissues of Mongolian and Han patients was lower than that in paracan-cerous and normal tissues,and the difference was statistically significant (all P 〈0.05).The expression of p14ARF in the paracancerous tissues was lower than that in the normal tissues,and the difference was statistically significant (P 〈0.05), Meanwhile,the expression of p14ARF itself in two nationalities had no significant difference (P 〉0.05).The positive rate of Hp was high in the gastric cancer tissues of patients in Mongolian and Han nationalities,and there was no significant differ-ence in the rate of infection between the two nationalities (P 〉0.05).The relative expression of p14ARF mRNA was statisti-cally significant among the cancer tissues,paracancerous tissues and normal tissues of Han patients with gastric cancer as well as in Mongolian patients (all P 〈0.05).There was no significant difference in the relative expression of p14ARF mR-NA of cancer tissues,paracancerous tissues and normal tissues of Mongolian and Han patients (P 〉0.05).Spearman cor-relation analysis showed that the positive expression of Hp and p14ARF was negatively correlated with each other in Han and Mongolian gastric cancer tissues (r =-0.406,-0.433,all P 〈0.05).Conclusions The expression of p14ARF is low, the positive rate of Hp is high,and p14ARF and Hp is negatively correlated in cancer tissues of patients with gastric cancer. The pathogenic mechanism of Hp and p14ARF in the Mongolian and Han patients with gastric cancer may be the same,and both of Hp and p14ARF may jointly promote the occurrence of gastric cancer.
