摘要
目的:通过建立1个超高效液相色谱-串联质谱法(UPLC-MS/MS)串联质谱分析方法,对大鼠脑组织中药物活性成分定量研究,探究芦丁对二苯乙烯苷透过血脑屏障的促进作用。方法:色谱条件:Kinetex C18色谱柱(100mm×4.6mm,1.8μm),Phenomenex C18保护柱;流动相:0.1%甲酸水(A)-乙腈(B)梯度洗脱,0-1min 90%A:1-3min 90%-30%A;3-4min 30%A;4-4.2min 30%-90%A;4.2-5.5min 90%A;流量:0.8m L/min;进样量:10μL。通过饲喂SD大鼠何首乌提取物与高、低剂量的芦丁配伍的药液,给药1h后乙醚麻醉,腹腔取血后,断头分离脑组织,取脑组织匀浆,经除杂处理后行UPLC-MS/MS分析。结果:何首乌芦丁配伍高、低剂量组大鼠脑组织中二苯乙烯苷含量高于何首乌组,且呈剂量依赖性。结论:芦丁配伍有助于提高二苯乙烯苷在脑组织中的浓度,芦丁对二苯乙烯苷透过血脑屏障具有促进作用,发挥中枢神经系统药效。
Objective: To quantitative study of active constituents in rat brain and explore the promoting effect of Rutin on stibene glucoside through blood-brain barrier by establishing UPLC-MS/MS tandem mass spectrometry. Methods: Rats received gavage administration with extracts of fleeceflower root combined with low and high dosages of Rutin respectively. 1h after administration rats were narcotized with ether, then abdominal blood was collected and brain homogenate was obtained. Rat brain homogenate was performed on UPLC-MS/MS using a Kinetex C18 column(100mm×4.6mm, 1.8μm) and Phenomenex C18 protection column. The mobile phases comprised acetonitrile and 0.1% formic acid. The analysis performed at a flow rate of 0.8 m L/min with the loading quantity as 10μL. Results: Rutin has a promoting effect on stilbene glycoside through the blood-brain barrier, and it helps to increase the concentration of stilbene glycoside in brain tissue to play the central nervous system efficacy.
