吡格列酮改善AD大鼠认知功能的研究 被引量:1

摘要 目的探讨吡格列酮(PIO)对阿尔茨海默病(AD)大鼠海马组织基因表达的影响。方法收录公众数据平台NCBI下基因芯片数据集GSE32536,采用R语言中的limma算法筛选差异表达基因,运用DAVID在线注释平台对差异基因进行功能分析。结果与对照组相比,PIO组大鼠海马组织中存在着44个差异表达基因(fold change≥1.5),包含40个上调基因,4个下调基因。40个上调基因属于胞外区细胞组分(GO-CC,P<0.01),它们主要参与了行为、运动行为、离子转运等生物学过程(GO-BP,P<0.01),发挥了底物通道活性、跨膜转运体活性等分子功能(GO-MF,P<0.01)。结论应用PIO能够改变AD大鼠的海马组织基因表达,促进大鼠的行为能力,改善认知功能。 Objective To explore the effect of pioglitazone(PIO) on gene expression profile in hippocampus at Alzheimer's disease(AD) mouse. Method The dataset GSE32536 was downloaded from NCBI database and analyzed by limma algorithm in R language, DAVID online tools was performed for gene function analysis. Results 44 differentially expressed genes(DEGs) were screened out in PIO group compared with control group, including 40 up-DEGs and 4 down-DEGs. The up-DEGs were mainly belonged to extracellular region component, involved in behavior-related biological process, acted the molecular function including channel activity and transporter activity. Conclusion Application of pioglitazone on AD mouse can cause genetic alterations in hippocampus, which may improve the behavioral outcome and cognitive function of AD mouse.
