摘要
目的建立一种基于病理切片定位石蜡包埋块上病变部位,针对病变部位进行准确取样,再进行超薄切片并进行透射电子显微镜(透射电镜)观察的方法。方法将人宫颈尖锐湿疣标本进行常规石蜡包埋、切片及HE染色,光学显微镜下观察并标记挖空细胞部位,再通过坐标纸确定石蜡包埋样本块上与切片上挖空细胞的对应部位,通过取样器获取石蜡块上目标部位,对其进行脱蜡及超薄切片,并进行透射电镜观察。结果透射电镜下观察到挖空细胞及人乳头状瘤病毒(HPV)样颗粒。结论建立了一种以病理切片定位石蜡包埋样本块上病变部位,对该部位进行准确取材并进行超薄切片透射电镜检测的方法。
Objective To establish ultrathin section technique which target pathologic cells based on the localization guidance of paraffin embedded section. Methods Cervical condyloma acuminatum samples were embedded with paraffin,sectioned and stained with hematoxylin and eosin( HE). Koilocytes were found on the HE stained section,and the counterpart site in paraffin embedded sample was localized with coordinate paper. The counterpart sample in paraffin block was picked out with dig tool and deparaffined. Then the sample was embedded with resin,ultrathin sectioned and observed under transmission electron microscope. Results Koilocytes and Human papillomavirus- like particles were founded on the ultrathin section with transmission electron microscope. Conclusion A method based on pathological section locating lesion on the paraffin embedded samples has been established to carry out the accurate sampling of the site and the detection of transmission electron microscope.
