色素上皮细胞衍生因子在膀胱癌旁黏膜及膀胱癌中的表达及其临床意义

The expression changes and clinical significance of PEDF in bladder mucosa and bladder cancer

目的 研究色素上皮细胞衍生因子（pigment epithelium derived factor,PEDF）在膀胱癌旁黏膜及膀胱中的表达变化及其意义.方法 收集在本院确诊为膀胱癌并且接受手术治疗患者的膀胱癌组织及癌旁组织,采用RT-PCR检测组织水平PEDF的mRNA表达差异,Western Blot及免疫组化IHC检测不同临床分期膀胱癌组织中PEDF的蛋白水平,并且比较了其同VEGF、MMP-2、MMP-9、MMP-12的相对表达.通过临床收集相应生理参数数据,分析PEDF同不同临床生理参数的相关性.结果 PEDF在膀胱癌旁黏膜中的表达水平无论是mRNA水平还是蛋白水平均显著高于膀胱癌组织（P＜0.05）,并且临床Ⅰ期和Ⅱ期的膀胱癌组织中PEDF的表达水平显著高于Ⅲ期、Ⅳ期（P＜0.05）,相关性分析表明,PEDF的阳性率同膀胱癌临床分期、淋巴结转移显著相关（P＜0.05）,同其他生理参数无显著相关性（P＞0.05）.结论 PEDF的表达差异同膀胱癌的发生和发展可能存在一定的相关性,具有一定的临床意义.

Objectives To investigate the expression changes and clinical significance of pigment epithelium derived factor （PEDF） in bladder mucosa and bladder cancer.Methods Bladder cancer tissue and tissue adjacent to carcinoma from patients in our hospital diagnosed with bladder cancer were collected,mRNA expression level of PEDF and protein levels in different clinical stages of bladder cancer tissue were tested by using RT-PCR,Western Blot and immunohistochemistry.The relevant expression level of VEGF 、MMP-2 、MMP-9 and MMP-12 were also analyzed by IHC.The relevance between PEDF and different physiological parameters was analyzed by collecting clinical data.Results mRNA level,protein expression level of PEDF in bladder cancer tissues were significantly lower than normal tissue.Correlation analysis showed that the positive rate of PEDF significantly associated with clinical stages,lymph node metastasis of bladder cancer （P 〈 0.05）,there was no significant correlation with other physical parameters （P 〉 0.05）.Conclusions There may be some relativity between PEDF expression differences and development of bladder cancer.
