摘要
目的建立Lewis肺腺癌小鼠移植瘤模型,取移植瘤进行EGFR基因测序,观察肺抑瘤膏对移植瘤的干预作用。方法使用C57BL/6小鼠,接种Lewis肺腺癌细胞,造模成功后,随机分4组,生理盐水对照组(组Ⅰ)、肺抑瘤膏组(组Ⅱ),3-MA组(组Ⅲ),Rap组(组Ⅳ),药物干预后,观察瘤重,并进行电镜、Western Blot检测及EGFR基因测序。结果 EGFR基因测序示:非突变型。肺抑瘤膏可以明显减轻移植瘤瘤重,组Ⅱ与生理盐水对照组瘤重比较,差异有统计学意义(P<0.05),组Ⅱ抑瘤率为25.22%;以自噬抑制剂3-MA、自噬诱导剂Rap作为对照分析,肺抑瘤膏组与3-MA组在瘤重、LC3/β-actin相对表达量两方面比较,差异均有统计学意义(P<0.05),而与Rap组在瘤重、LC3/β-actin相对表达量两方面比较,差异均无统计学意义(P>0.05)。电镜提示肺抑瘤膏可影响肿瘤细胞自噬。结论肺抑瘤膏能够明显抑制EGFR非基因突变型肺腺癌移植瘤的生长,且可能通过影响肿瘤细胞自噬起到治疗作用。
Objective Through the establishment of Lewis lung tumor model in mice, the transplanted tumors were detected EGFR gene sequencing and Anti-lung Tumor Ointment were observed the intervention effect for transplanted tumors. Methods The C57 BL / 6mice were inoculated with Lewis lung carcinoma cells to established of Lewis lung tumor model. The mice were randomly divided into four groups after successful modeling. Normal saline group(group I), Anti-lung Tumor Ointment group(group II), 3-MA group(group III), Rap group(group IV). We observed the tumor weight and the electron microscope, and Western Blot, and EGFR gene sequencing. Results EGFR gene sequencing shows: Non-mutant. Anti-lung Tumor Ointment can significantly reduce the tumor weight. GroupⅡcompared with groupⅠin tumor weight, expression were different(P〈0.05).Group II tumor suppressor rate was 25.22%. GroupⅡcompared with group III in tumor weight and LC3/β-actin relative expression, expression were different(P〈0.05).GroupⅡcompared with group IV in tumor weight and LC3/β-actin relative expression, expression were no different(P〉0.05). Electron microscopy result show that Anti-lung Tumor Ointment can affect autophagy. Conclusion Anti-lung Tumor Ointment can significantly inhibit transplanted tumor growth of EGFR non genetic mutations of lung adenocarcinoma in lewis lung model mice, and may play a therapeutic rolethrough the induction of autophagy.
