4-N-亚硝基甲基氨-1-(3-吡啶基)丁酮对肺腺癌A549细胞增殖、凋亡的影响

摘要 目的研究4-N-亚硝基甲基氨-1-(3-吡啶基)丁酮对3_549细胞增殖、凋亡的影响。方法用含3%4-N-亚硝基甲基氨-1-(3-吡啶基)丁酮的培养基处理肺腺癌A549细胞,分别收集培养1周、2周、3周的细胞,采用四甲基偶氮唑盐法检测细胞增殖特性,Hoeehst33258荧光染色法观察细胞凋亡。结果对照组A549细胞培养3周后吸光度差异未见统计学意义(F=0.554,P=0.650),经4-N-亚硝基甲基氨.1.(3-吡啶基)丁酮处理1周、2周、3周后A549细胞吸光度分别为(0.516±0.007)、(0.485±0.005)、(0.432±0.013),差异有统计学意义(F=171.804,P〈0.001),各处理组与对照组吸光度比较差异有统计学意义(P均〈0.001)。随着4-N.亚硝基甲基氨-1-(3-吡啶基)丁酮对A549细胞作用时间的增加,增殖抑制率逐渐增加,分别为(22.28±1.58)%、(27.06±0.70)%、(34.60±2.18)%,差异有统计学意义(F=169.77,P〈0.001)。4.N.亚硝基甲基氨.1-(3-吡啶基)丁酮可诱导A549细胞的凋亡,随着作用时间的延长,各处理组细胞凋亡率逐渐增加,分别为(16.32±0.93)%、(22.92±1.11)%、(29.88±0.81)%,与对照组[(10.22±1.29)%]相比,差异有统计学意义(F=387.171,P〈0.001)。结论4-N-亚硝基甲基氨-1-(3-吡啶基)丁酮可抑制A549细胞的增殖,并促进其凋亡。 Objective To study the effect of 4-N-methylamine-nitroso-l-(3-pyridyl) acetone on the proliferation, apoptosis of lung adenocarcinoma A549 cell. Methods Lung adenocarcinoma A549 cells were treated by medium containing 3% 4-N-methylamine-nitroso-l-(3-pyridyl) acetone. A549 cells cultured for 1 weeks, 2 weeks, 3 weeks were collected separately, and the cell proliferation was detected by MTF method, and the cell apoptosis was observed by Hoechst 33258 fluorescence staining. Results There was no significant difference in the absorbance of A549 ceils cultured for 3 weeks in con- trol group ( F = 0. 554, P = 0. 650 ). After the 4-N-methylamine-nitroso-1- (3-pyridyl) acetone treated for one week, two weeks and three weeks , the absorbance of A549 cells were 0. 516 ± 0. 007, 0. 485 ± 0. 005, 0. 432 ± 0. 013, the differences were significant (F = 171. 804, P 〈 0. 001 ). There were signifi- cant differences in the absorbance between each treatment group and control group (P all 〈 0. 001 ). With the acting time of 4-N-methylamine-nitroso-l-(3-pyridyl) acetone on A549 cells increased, the prolifera- tion inhibition rate gradually increased, respectively was ( 22. 28 ± 1.58 ) %, ( 27.06 ± 0. 70 ) %, (34. 60 ± 2. 18 ) %, the difference were significant ( F = 169.77, P 〈 0. 001 ). 4-N-methylamine-nitroso- 1- (3-pyridyl) acetone induced apoptosis of A549 cells, with the extend of acting time, the apoptosis rate in each treatment group gradually increased, which were ( 16. 32±0. 93 ) %, ( 22. 92 ± 1.11 ) % and (29. 88 ±0. 81 ) %, compared with the control group [ (10. 22 ± 1.29)% ], the differences were signifi- cant ( F = 387. 171, P 〈 0. 001 ). Conclusions 4-N-methylamine-nitroso-l-(3-pyridyl) acetone can inhibit proliferation and promote the apoptosis of A549 cells.
