替丁类药物与全肠外营养液配伍后抗溃疡作用的研究 被引量:3

The effect of H_2 receptor antagonist mixed with total parenteral nutrient on gastric ulcer in rat model

摘要 目的考察替丁类药物与全肠外营养液(total parenteral nutrient,TPN)配伍后的体外稳定性及抗溃疡作用。方法根据临床处方配比,将替丁类药物加入到全肠外营养液中,得到不同浓度的含药制剂及对照制剂,配伍液在室温下放置24h,于不同时间点观察、测定和记录配伍液的外观性状、脂肪乳粒径大小及分布、pH值和渗透浓度;建立组胺致小鼠溃疡模型,所得数据采用SPSS统计分析软件进行t检验,观察西咪替丁(cimetidine,CMT)、法莫替丁(famotidine,FMT)与TPN配伍制剂对胃溃疡的抑制作用。结果替丁类药物与全肠外营养液配伍后,制剂的外观、脂肪乳粒径大小及分布、pH值和渗透浓度在24h内均没有发生显著性变化;药效学实验中,CMT-TPN(50.0、100.0 mg/kg)和FMT-TPN(4.2、8.4mg/kg)均可显著抑制组胺致小鼠溃疡的发生,其溃疡抑制率分别为49.44%、60.69%和53.52%、67.29%。结论替丁类药物与TPN配伍后能够稳定24h,且有显著预防和治疗胃溃疡的作用,本研究为其临床配伍使用提供了数据支持。 Objective To observe the effect of H2 receptor antagonist mixed with total parenteral nutrient(TPN)on experimental gastric ulcer. Methods H2 receptor antagonist was mixed with TPN at different concentration and the mixtures were kept at room temperature for 24 h.The appearances of the mixtures were observed at different time points.At the same time,the change of pH value,osmolality and the accumulation of fat emulsion drops were observed and recorded.Rat model with gastric ulcer was induced by histamine.The inhibitory effect of Cimetidine(CMT)and Famotidine(FMT)mixed with TPN were observed,respectively. Results There were no significant changes in the appearance,pH value,osmolality,the size and distribution of accumulated fat emulsion drops of the mixtures.CMT-TPN(50.00 mg/kg,100.00 mg/kg)showed significant protective effect on experimental gastric ulcers.The ulcer inhibition rate was 49.44% and 60.69%,respectively.FMT-TPN(4.2,8.4mg/kg)showed significant protective effect on experimental gastric ulcers.The ulcer inhibition rate was 53.52%and 67.29%,respectively. Conclusions Cimetidine-injection mixed in TPN is stable at room temperature for 24 h.CMT and FMT may have protective and therapeutic effect on gastric ulcer in rat model.
