摘要
目的观察异甘草素对大鼠脑胶质瘤组织单核细胞趋化蛋白-1(MCP-1)和白介素-1(IL-17)表达的影响,探讨异甘草素抗胶质瘤作用机制。方法方法清洁级Wistar雌性大鼠,n=36只,随机分为3组:假手术组(n=12),模型组(n=12)与异甘草素组(n=12)。异甘草素组腹腔注射异甘草素(500μg/kg/d),模型组腹腔注射等量生理盐水,连续给药一周后处死大鼠。免疫组织化学和Western blot方法检测肿瘤组织MCP-1和IL-17蛋白表达水平的变化。应用CCK-8试剂盒检测异甘草素对人U87胶质瘤细胞增殖能力的影响。结果与假手术组相比,模型组与异甘草素组大鼠脑组织MCP-1和IL-17的蛋白表达的平均光密度值显著增加(P<0.01),而异甘草素组显著低于模型组(P<0.01)。与对照组相比,异甘草素显著抑制了人U87胶质瘤细胞增殖。结论异甘草素抑制人U87胶质瘤细胞增殖可能与降低脑胶质瘤大鼠MCP-1和IL-17的表达相关。
Objective To investigate the effect of isoliquiritigenin on the expressions of monocyte chemoattractant protein-l(MCP-1) and interlukin-17(IL-17) in C6 glioma rat models and to explore the antiglioma mechanism. Methods 36 Wistar female rats were randomly divided into 3 group: sham group(n=12)and model gronp(n=12,intraperitoneal injection of normal saline) and isoliquiritigenin group(n=12,intraperitoneal injection of isoliquiritigenin of 500 blg/kg/d). The expressions of MCP-1 and IL-17 were detected by immunohistochemistry and Western blot, and CCK-8 kit was applied to detect the proliferation of U87 glioma cells. Results The mean optic density(MOD) values of MCP-1 and IL-17 positive expressions in brain tissues were higher significantly in model group than in sham group(P〈0.01) , but lower in isoliquiritigenin group than in model group(P〈0.01). Compared with control group, isoliquiritigenin significantly inhibited the proliferation of U87 cells, Conclusion The inhibition effect of isoliquiritigenin on glioma cells might be related to the downregulation of MCP-1 and IL-17 expression in rats with brain glioma.
