Expression and clinical significance of miR-196a/b in non small cell lung cancer

摘要 目的分析miR-196a和miR-196b在非小细胞肺癌中的表达及其临床意义。方法收集非小细胞肺癌手术标本157例及癌旁组织标本50例,提取总RNA,采用Realtime PCR方法检测miR-196a和miR-196b在非小细胞肺癌及其癌旁组织中的表达,分析二者表达的相关性,并分析其表达与临床病理特征及其预后的关系。结果 miR-196a和miR-196b在非小细胞肺癌组织中的表达高于癌旁肺组织,二者表达呈正相关(r=0.457,P<0.001)。miR-196a和miR-196b联合高表达与组织类型(P=0.010)、淋巴结有无转移(P=0.016)、远处转移(P=0.007)及临床分期(P=0.007)相关,而与年龄、性别、组织分化程度无显著相关性(P>0.05)。Kaplan-Meier生存分析显示miR-196a和miR-196b联合高表达组患者生存期显著低于联合低表达或者单独高表达组(P<0.001),Cox风险比例模型分析显示miR-196a和miR-196b联合高表达为非小细胞肺癌患者独立的危险因素。结论 miR-196a和miR-196b在非小细胞肺癌中异常高表达可能是潜在的肺癌预后分子标志物。 Objective To analyze the expression and clinical significance of miR-196a and miR-196b in non small cell lung cancer(NSCLC). Methods 157 cases surgical specimens of NSCLC patients and 50 cases adjacent normal tissue were collected. Total RNA was extracted, miR-196a and miR-196b expressions were detected by real-time PCR, correlation of miR-196a and miR-196b expression with elinieopathologieal characteristics and prognosis was analyzed. Results The expression levels of miR-196a and miR-196b were higher in NSCLC than in adjacent normal lung tissue, and positively correlated to each other(r=0.457, P〈0.001), and positively correlated to histological type ( p =0.010), lymph node metastasis ( p =0.016), distant metastasis ( P =0.1307) and clinical staging ( p = 0.007), but not for age, sex and the degree of histological differentiation(P 〉0.05). The overall survival time of patients was significantly lower in both miR-196a and miR- 196b high expression than in both low expression or single high expression group(P 〈0.001), and both high miR-196a and miR-196b expression were the independent risk factor for NSCLC patients. Conclusion The high expression of miR-196a and miR-196b might be a potential marker for predicting the prognosis of NSCLC.
