基于NF-κB信号通路倍半萜烯内脂化合物对牙周病破骨细胞活化因子作用的研究 被引量:2

Study on the effect of sesquiterpene lactone compounds on osteoclast activating factor IL-1 in periodontal disease based on NF-κB signaling pathway

摘要 目的:探究基于NF-κB信号通路倍半萜烯内脂化合物对牙周病破骨细胞活化因子作用的研究。方法:选取2周龄的牙周病大鼠动物模型15只为观察组,2周龄的健康大鼠6只为对照组。观察组实验大鼠随机分为3组:观察组1实验大鼠注射小白菊内酯(PAR)2.5 mg/kg/d;观察组2注射粘毛鼠尾草活性单体2 mg/kg/d;观察组3及对照组每天注射等量的生理盐水。通过荧光定量PCR技术、酶联免疫反应(ELISA)等方法对观察组与对照组牙周膜成纤维细胞(periodontal ligament fibroblasts,PDLF)中基于NF-κB信号通路的牙周病破骨细胞活化因子(IL-1β)mRNA和蛋白表达进行测定,并观察实验动物模型牙周病的治疗状况。结果:与对照组(1.02±0.08,3.2±0.1)ng相比,观察组1(1.1±0.11,3.4±0.15)ng、观察组2(1.12±0.05,3.36±0.13)ng大鼠牙周组织中NF-κB以及IL-1β含量的差异均无统计学意义(P>0.05)。NF-κB以及IL-1β含量在观察组3(3.92±0.03,8.59±0.16)ng中与在观察组1、观察组2中的差异均有统计学意义(P<0.05),并且治疗2周后发现经倍半萜烯内脂化合物(小白菊内酯、粘毛鼠尾草活性单体)治疗的观察组1与观察组2中牙周状况均得以改善。结论:倍半萜烯内脂化合物可以通过NF-κB信号通路作用于牙周病破骨细胞活化因子(IL-1β)进而改善牙周病状况。 Objective: To study the effect of sesquiterpene lactone compounds on osteoclast activating factor IL-1 in h PDLF periodontal disease based on NF-κB signaling pathway. Method: Fifteen 2-weeks-old rats with periodontal disease were selected as the observation group,and 6 healthy 2-weeks-old rats were selected as the control group. The observation group's rats were randomly divided into 3 groups. The rats were injected 2. 5 mg / kg / d parthenolide( PAR) in group 1,injected2 mg / kg / d salvia roborowskii active monomer in group 2,and injected daily with the same amount of saline in group 3 and the control group. Osteoclast activating factor IL-1 mRNA and protein expression were measured by fluorescence quantitative polymerase chain reaction( PCR) and enzyme linked immune assay( ELISA) method of periodontal ligament fibroblasts( PDLF) based on NF-κB signaling pathway in periodontal disease. And the treatment of periodontal disease in the observation group was observed. Results: Compared with the control group( 1. 02 ± 0. 08,3. 2 ± 0. 1) ng,there were no statistically significant difference in the NF-κB and IL-1 content of rats periodontal tissue both in group 1( 1. 1 ± 0. 11,3. 4 ± 0. 15) ng and group 2( 1. 12 ± 0. 05,3. 36 ± 0. 13) ng,respectively( P 0. 05). The content of NF-κB and IL-1 in group 3( 3. 92 ± 0. 03,8.59 ± 0. 16) ng when compared with group 1 and group 2 respectively,there were statistically significant differences( P 0.05). After 2 weeks of the treatment by sesquiterpene lactone compounds including parthenolide and salvia roborowskii active monomer both in group 1 and group 2,the periodontal conditions have been improved. Conclusion: The treatment effect of sesquiterpene lactone compounds on osteoclast activating factor IL-1 in periodontal disease based on NF-κB signaling pathway have been proved.
