摘要
刺参(Apostichopus japonicus)利用夏眠的习性来应对夏季的高温胁迫,长期的代谢减退和基因转录抑制是刺参夏眠调控过程中的关键策略。本研究克隆分析了刺参体内关键组蛋白脱乙酰基转移酶(HDACs)的基因全长及结构特征,测定了该家族成员(HDAC1,HDAC3和HDAC4)在夏眠期间的基因表达模式。研究表明:刺参HDAC3和HDAC4与紫海胆(Strongylocehtrotus purpturatus)相关基因的同源性最高,分属HDACs家族的Ⅰ型和Ⅱ型。定量表达分析表明,刺参不同组织中同属Ⅰ型的HDAC1和HDAC3基因在刺参夏眠不同阶段的表达均无显著差异;而属于Ⅱ型的HDAC4基因在深度夏眠阶段表达量显著上调。研究结果表明,II型HDACs在刺参夏眠基因转录抑制中具有潜在的特殊调节作用。
Sea cucumber,ApostichopusjaponicusSelenka,survives high summer temperature by entering into aestivation which is characterized by hypometabolism and global transcriptional depression.We investigated the hypothesis that aestivationis associated with acetylation-dependent epigenetic mechanisms by cloning,sequencing and quantifying the transcripts of important members of histone deacetylases(HDACs)family.The results showed that the deduced amino acid sequences and characteristic motifs of sea cucumber HDAC3 and HDAC4were highly similar to those of their echinoderms.No significantly variation was observed in HDAC1 and HDAC3belonging to class I in different tissues of sea cucumber during aestivation.However,the expression of HDAC4 belonging to class II was significantly up-regulated in intestine and muscle during deep-aestivation.The findings in present study suggested that class II HDACs involved in transcriptional silencing during aestivation.
