摘要
目的:建立胡颓子叶中银锻苷含量测定的方法。方法:采用80%甲醇提取样品;HPLC法测定,色谱条件为:色谱柱为Agilent C_(18)(250 mm×4.6 mm,5μm),流动相:乙腈-0.1%磷酸水溶液,梯度洗脱,柱温:30℃,流速:1.0 ml·min^(-1),检测波长:315 nm,进样量:20μl。结果:银锻苷在0.01~0.20 mg·ml^(-1)范围内与峰面积积分值呈良好的线性关系(r=0.999 9),平均回收率为99.14%,RSD为1.33%(n=6)。结论:该方法简便、快速、准确、重复性好,可用于胡颓子叶的含量测定。
Objective: To establish an HPLC method for the determination of tiliroside in Elaeagnus pungens leaves. Methods: The samples were extracted by 80% methanol. The chromatographic separation was carried out on an Agilent C18 column (250 mm×4.6 mm, 5 μm) with the mobile phase consisting of acetonitrile-0.1% phosphoric acid with gradient elution at a flow rate of 1.0 ml ·min^-1. The detection wavelength was set at 315 nm, the injection volume was 201xl and the column temperature was 30~C. Results: The linear range of tiliroside was 0.01-0.20 mg·ml ^- 1 ( r = 0. 999 9) with the average recovery of 99.14% ( RSD = 1.33 %, n = 6). Conclusion: The method is simple, accurate and reproducible, which is suitable for the content determination of E. pungens leaves.
