芒果苷联合葛根素对自发性高血压大鼠肾脏炎性损伤的协同保护作用

Synergistic Protective Effect of Mangiferin Joint Puerarin on the Renal Inflammatory Injury in Spontaneously Hypertensive Rats

以自发性高血压大鼠作为实验动物模型,观察芒果苷联合葛根素对SHR形态学及炎症因子IL-6,IL-10,TNF-α表达的影响,探讨芒果苷联合葛根素是否能增强保护肾脏炎症损伤的作用。将8只WKY大鼠作为对照组A,再将72只SHR大鼠随机分为模型组B、苯那普利组C(10 mg·kg^(-1)·d^(-1))、芒果苷组D(20 mg·kg^(-1)·d^(-1))、葛根素组E(20 mg·kg^(-1)·d^(-1))、芒果苷(20 mg·kg^(-1)·d^(-1))+葛根素片(10 mg·kg^(-1)·d^(-1))组F、芒果苷(20 mg·kg^(-1)·d^(-1))+葛根素片(20 mg·kg^(-1)·d^(-1))组G、芒果苷(20 mg·kg^(-1)·d^(-1))+葛根素片(40 mg·kg^(-1)·d^(-1))组H、葛根素片(20 mg·kg^(-1)·d^(-1))+芒果苷(10 mg·kg^(-1)·d^(-1))组I、葛根素片(20 mg·kg^(-1)·d^(-1))+芒果苷(40 mg·kg^(-1)·d^(-1))组J。每天给药10 m L/kg连续灌胃,灌胃给药2个月后停药,杀鼠前禁食12 h。用显微镜观察大鼠肾组织病理形态结构,免疫组化法检测大鼠肾组织IL-6、IL-10、TNF-α的表达,ELISA法检测SHR IL-6、IL-10、TNF-a水平。各组大鼠肾脏组织均未见明显病理形态学改变;免疫组化检测和酶联免疫检测结果显示:与模型组比较,各组大鼠肾组织IL-6,IL-10,TNF-α含量均有不同程度下降。芒果苷联合葛根素对自发性高血压大鼠肾脏组织形态学无显著影响;可下调大鼠肾脏组织异常升高的IL-6,IL-10,TN-α水平。

In spontaneously hypertensive rats as the experimental animal model,observation of mangiferin and puerarin on SHR morphology and the influence of inflammatory factors IL-6,IL-10,TNF- expression,study of mangiferin and puerarin whether can enhance protection of target organ inflammatory injury in rats. 8 male WKY rats as control group A,in 72 only 10 weeks of SHR rats as the research object,were randomly divided into model group B,positive control C（ benzene that split）（ 10 mg·kg^-1·d^-1）,mangiferin in group D（ 20 mg·kg^-1·d^-1）,puerarin alone group of Group E（ 20 mg · kg^-1·d^-1）,mangiferin（ 20 mg·kg^-1·d^-1） ＋ puerarin tablet（ 10mg·kg^-1·d^-1） in group F,mangiferin（ 20 mg·kg^-1·d^-1） ＋ puerarin tablet（ 20 mg·kg^-1·d^-1） in group G,mangiferin（ 20 mg·kg^-1·d^-1） ＋ puerarin tablet（ 40 mg·kg^-1·d^-1） in group H,puerarin tablet（ 20 mg·kg^-1·d^-1） ＋ mangiferin（ 10 mg·kg^-1·d^-1） in group I,puerarin tablet（ 20 mg·kg^-1·d^-1） ＋ mangiferin（ 40 mg · kg^-1· d^-1） in group J. lavage for 2 months after drug withdrawal,kill the rats of fasting 12 h. Morphology and structure of rat renal tissue pathological observation by microscope,the expression of IL-6,IL-10 and TNF- in renal tissue of SHR was detected by immunohistochemistry method,Detection of SHRIL-6,IL-10 and TNF-a levels by ELISA method. The kidney tissues of rats in each group showed no obvious pathological changes;the detection results of ELISA and immunohistochemical detection of enzyme display： compared with model group,the contents of IL-6,IL-10 and TNF- in myocardium of rats in each group were decreased in different degrees.Mangiferin combined with Puerarin had no significant effect on the morphology of kidney in spontaneously hypertensive rats; can downregulate rat kidney muscle abnormal increase of IL-6,IL-10,TNF- a level.
