摘要 目的应用生物信息学技术,全面了解人谷胱甘肽过氧化物酶(Human Glutathione Peroxidase,h GPx)蛋白家族的理化性质、结构及功能.方法以NCBI中报道的h GPx家族8个成员为研究对象,利用Prot Param,Net NGlyc,Net OGlyc,Net Phos,SOPMA,Clustal W,Pymol,MEGA 5.05等多种生物信息学软件,分别对其分子量、等电点、亲水性、糖基化、磷酸化和结构等方面进行分析,并建立其分子系统进化树.结果通过比对发现:h GPx超家族8个成员的物理性质存在明显差异,而在结构及功能上存在相似特征.结论 h GPx是一类具有明确酶活性的硒蛋白,具有清除体内过量的ROS、保护机体免受氧化损伤的作用. Objective To comprehensively understand the physicochemical properties,structures and properties of human glutathione peroxidase (hGPx) family by bioinforrnatics analysis. Method The eight members of hGPx family were used as the research objects. Bioinforrnatics software like ProtParam, NetNGlyc, NetOGlyc, NetPhos, SOPMA, Clustal W, Pymol and MEGA 5. 05 were applied to analyze the molecular weight, PI, hydrophilic, glycosylation,phosphorylation and structure, And an evolutionary tree of hGPx was built in the study. Results It was found that members of hGPx family had an obvious difference in their physical properties. Besides, there are similar characteristics in their structures and functions. Conclusion hGPx is a kind of selenoprotein with definite enzyme activities,and can scavenge the excessive ROS in the body and protect the body from being damaged by oxidation.
