不同方法制备新鲜冰冻血浆对冷沉淀凝血因子质量的影响 被引量:13

摘要 目的:分析不同方法制备新鲜冰冻血浆对冷沉淀凝血因子中纤维蛋白原(Fg)和凝血因子Ⅷ(FⅧ)含量的影响。方法随机选取90份本中心2014年3-6月自愿献血者400 ml全血,按照不同制备流程分成3组,第1组全血滤除白细胞过滤新鲜冰冻血浆(fresh frozen plasma,FFP),经病毒灭活后制备冷沉淀因子;第2组全血分离FFP,经病毒灭活后制备冷沉淀因子;第3组全血白膜法制备浓缩血小板分离FFP,经病毒灭活后制备冷沉淀因子。采用水浴融化法,随机抽取冷沉淀90份,取小辫5-10 cm,经37℃水浴融化后留取标本2-3 ml,测定Fg,FⅧ含量并进行对比分析。结果3组中Fg (mg/袋,200 ml FFP制备)含量分别为:161.3±29.6,168.5±33.9,156.6±36.5;FⅧ(IU/袋,200 ml FFP制备)为122.4±34.9,131.5±30.2,101.9±21.7;抽检3组冷沉淀凝血因子中Fg、FⅧ含量合格率分别为91.2%、94.3%、80.9%,均达到要求。抽检结果显示第2组Fg 、FⅧ含量最高,抽检合格率达94.3%;3组质量抽检合格率由高到低依次是第2组>1组>3组;3种制备方法组间Fg 、FⅧ含量比较,差异无统计学意义(P>0.05)。结论3种不同方法制备新鲜冰冻血浆,冷沉淀凝血因子质量指标均符合国家标准。
