活血通脉颗粒对兔颈动脉粥样硬化干预作用的实验研究 被引量:2

摘要 目的观察活血通脉颗粒对兔颈动脉粥样硬化(CAS)模型中颈动脉病变的影响。方法将96只清洁级新西兰大白兔随机抽取10只为正常对照组,其余86只喂高脂饲料,11周后造模成后56只随机分为模型组、自然消退组、普通饲料活血通脉颗粒低、中、高剂量组、普通饲料辛伐他汀组、高脂饲料活血通脉颗粒中剂量组、高脂饲料辛伐他汀组8组,分别连续给药5周,各组经耳缘静脉采血,全自动生化分析仪测定血清总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)。取兔颈动脉组织行HE染色,观察其病理改变。结果正常组颈动脉内皮光滑,未见斑块和脂质沉积,模型组内皮明显增生,斑块内含有大量泡沫细胞,粥样斑块分布广泛。普通饲料活血通脉颗粒组较模型组内皮增生程度明显减轻,斑块内含较少泡沫细胞。普通饲料高剂量组整个动脉管壁层次尚清晰。模型组TC、TG、HDL-C、LDL-C与正常组比较均有明显升高(P<0.01)。给药5周后,活血通脉颗粒低、中、高剂量组和辛伐他汀组血脂4项与自然消退组比较,明显下降(P<0.05);普通饲料活血通脉颗粒中、高剂量组和辛伐他汀组与活血通脉颗粒低剂量组比较,明显下降(P<0.05);普通饲料活血通脉颗粒高剂量组与普通饲料辛伐他汀比较,差异无统计学意义(P>0.05)。结论活血通脉颗粒可能抑制颈动脉粥样硬化形成的作用。 Objective: To discuss the intervention effect of Huoxue Tongmai granule on carotid arteriosclerosis (CAS) in rabbits and the curative effect of lipid regulation,stable plaques in science evaluation,thus providing theoretical basis for clinical medication. Methods: 10 rabbits were randomly selected from 96 New Zealand white rabbits,clean level only,as the normal control group,the remaining 86 were fed with high fat forage. 11 weeks later, 56 model rabbits were randomly divided into model group, natural healing group and normal forage Huoxue Tongmai granule low, medium and high dose group, normal forage simvastatin group, high-fat diet Huoxue Tong- mai granule medium dose group,high-fat feedstuff simvastatin group,totally 8 groups with 7 rabbits in each group. After each group was treated continuously for five weeks,blood was collected from ear marginal veins,TC, TG,HDL-C, LDL-C were detected with automatic biochemical analyzer. The rabbit carotid artery tissue was ex- tracted for HE staining to observe its pathologic change. Results: The carotid artery endothelium was smooth without plaques and lipid in the normal group. In the model group,endothelial proliferation was obvious,plaque contained a large number of foam cells,atheromatous plaques were widely distributed. The endothelial prolifera- tion condition was significant improved and plaque contained less number of foam cells in the normal forage Huoxue Tongmai granule group. The whole arterial wall level was relatively clear in the normal forage Huoxue Tongmai granule high dose group. Compared with normal group,TC,TG,I-IDL-C,LDL-C were significantly increased in the model group(P〈0.01 ). In five weeks after the treatment,Compared with the natural healing group,TC,TG,HDL-C,LDL-C were significantly decreased in the Huoxue Tongmai granule low,medium and high dose group and simvastatin group (P 〈 0.05). Compared with the Huoxue Tongmai granule low dose group,TC,TG, HDL-C,LDL-C were significantly decreased in the normal forage Huoxue Tongmai granule medium and high dose group and simvastatin group (P 〈 0.05). There was no statistical difference between the normal forage Huoxue Tongmai granule high dose group and normal forage simvastatin group (P〉 0.05). Conclusion: Huoxue Tongmai granule can inhibit the carotid artery atherosclerosis formation.
