非达霉素的菌种选育及发酵工艺优化 被引量:3

Production of Fidaxomicin by Strain Breeding and Optimization of Its Fermentation Process

摘要 对产非达霉素的出发菌株桔橙指孢囊菌(Dactylosporangium aurantiacum)-18进行紫外(UV)和亚硝基胍(NTG)诱变,并通过链霉素抗性模型筛选,获得突变菌株D.aurantiacum N-61,非达霉素摇瓶发酵产量为1 141 mg/L。进一步优化该菌株在5 L发酵罐上的培养工艺,通过调整发酵培养基处方(g/L),在乳糖75.0、甘油20.0、棉籽饼粉10.0、酵母粉10.0、CaCO_3 4.0和MgSO_4·7H_2O 2.0的条件下,非达霉素产量达到3 343 mg/L。 Dactylosporangium aurantiacum-18, a fidaxomicin producing strain, was mutated with UV and NTG and then screened in the plate with streptomycin. As a result, a mutant D. aurantiacum N-61 was obtained with the fidaxomicin titer of 1 141 mg/L in the shake flask. Furthermore, the fermentation process was investigated by optimizing medium composition in a 5 L fermentator. In the medium (g/L) containing lactose 75.0, glycerol 20.0, cottonseed powder 10.0, dry yeast 10.0, CaCO3 4.0, and MgSO4·7H2O 2.0, the yield of fidaxomicin could reach 3 343 mg/L.
