摘要
介绍了一种免标记质谱分子成像新技术,即红外激光解析/真空紫外激光后电离质谱成像方法.该方法具有样品前处理简单,无需人为添加基质等优点.同时,这种成像方法还可以通过在时间和空间上独立优化2束激光的波长、强度以及延迟时间来获得最佳的质谱成像探测条件.首次应用该实验技术成功地探测抗癌药物吖啶黄素在小鼠肾脏组织切片中的空间分布,其空间分辨率低于100μm,单一成像点检测灵敏度可达0.1 pmol.这种新型的质谱成像技术在临床医学和药物开发领域具有很好的应用前景.
A new mass imaging technique for biological tissues using laser desorption postionization mass spectrometry is proposed. This technique does not require multiple sample preparation and avoids the spectra interferences. Moreover,this imaging method can be used to optimize wavelength independently of two laser beams in time and space.It also can adjust the delay time between the desorption laser and the ionization laser. The first application of this experimental technique was applied successfully to detect the spatial distribution of anticancer drug,c14h15n3cl2,in mouse kidney tissues. This helps us get the best mass spectra and images with a resolution of 100 μm and a limit of 0.1 pmol. All the results show that this new imaging technique is promising in the areas of clinical medicine and medicinal development in the future.
