摘要 目的:采用高效液相色谱(HPLC)法建立一种同时测定金英黄归汤中芦丁、槲皮素和木犀草素的含量。方法:采用Hypersil ODS2 C18色谱柱(150 mm×4.6 mm,5μm);以甲醇-0.4%磷酸(40∶60)为流动相;检测波长为360 nm;柱温为40℃和;流速为0.8 m L·min^-1。结果:芦丁、槲皮素和木犀草素分别在350.00μg·mL^-1-21.88μg·mL^-1(R^2=0.999 8)、245.00μg·mL^-1-15.31μg·mL^-1(R^2=0.999 9)和240.00μg·mL^-1-15.00μg·mL^-1(R^2=0.999 9)范围内线性关系良好。中间精密度试验RSD分别为1.05%、1.16%和0.50%;稳定性试验RSD分别为2.00、%1.79%和1.60%;重复性试验RSD分别为1.78%、1.23%和1.84%;平均回收率分别为芦丁99.08%(RSD=0.81%),槲皮素100.5%(RSD=1.82%),木犀草素99.92(RSD=1.19%)。结论:建立的该测定方法简便快捷,准确可靠,重复性好,可用于同时测定金英黄归汤中芦丁、槲皮素和木犀草素的含量测定。 Objective:To establish the simultaneous determination methods of rutin,quercetin and mignonette in Jinying Huanggui Decoction by HPLC.Method: Hypersil ODS2 C18column(150 mm × 4.6 m,5 μm) was used.The mobile phase was methanol-0.4% phosphoric acid solution(40 ∶ 60).The detection wavelength was set at 360 nm.The column temperature was 40 ℃.The flow rate was 0.8 m L·min^-1.Results:The linear ranges of rutin,quercetin and mignonette were 350.00 μg·mL^-1- 21.88 μg·mL^-1(R^2= 0.999 8),245.00 μg·mL^-1- 15.31 μg·mL^-1(R^2= 0.999 9)and 240.00 μg·mL^-1- 15.00 μg·mL^-1(R^2= 0.999 9) respectively.The RSD values of intermediate precision test were 1.05%,1.16% and 0.50% respectively.The RSD values of stability test were 2.00%,1.79% and 1.60% respectively.The repeatability test results were 1.78%,1.23% and 1.84% respectively.The average recoveries of rutin,quercetin and mignonette were 99.08%(RSD = 0.81%),100.5%(RSD = 1.82%) and 99.92(RSD = 1.19%) respectively.Conclusion This method could be simple,accurate,repeatable and suitable for the simultaneous determinations of rutin,quercetin and mignonette in Jinying Huanggui Decoction.
