柴胡皂苷-D抑制宫颈癌Hela细胞的分子机制研究 被引量:20

摘要 目的:宫颈癌是女性常见恶性肿瘤之一,严重影响女性健康。柴胡皂苷是中药柴胡抗炎、抗肿瘤等作用的主要成分,其中柴胡皂苷D效果尤为明显。研究探讨柴胡皂苷D对宫颈癌细胞的作用的分子机制。方法:采用CCK-8法检测柴胡皂苷D对Hela细胞增殖的影响,利用流式细胞仪检测柴胡皂苷影响Hela细胞凋亡和细胞周期,同时利用定量PCR法检测细胞周期及凋亡相关基因的表达水平。结果:与对照相比,柴胡皂苷D能够显著抑制Hela细胞的增殖(P<0.01),通过流式细胞检测,发现柴胡皂苷D能够增加细胞凋亡比例(P<0.05),将Hela细胞阻滞在细胞周期G1期。进一步实验发现,柴胡皂苷D能够降低周期素Cyclin D1和Cyclin E,进而影响细胞周期;同时增加凋亡促进基因Bax表达,降低凋亡抑制基因Bcl-2基因表达,从而影响细胞凋亡。结论:柴胡皂苷D通过周期蛋白阻滞Hela细胞周期停滞在G1期,通过影响凋亡相关蛋白诱导Hela细胞凋亡。 Objective: Cervical cancer is one of common female cancers and harmful to health of women in the world.Saikosaponin is the main component in Radix Bupleuri which has anti-inflammation and anti-cancer effects.Saikosaponin-d(SSD) is the most effective member of Saikosaponin.At present,we explored the function and mechanism of SSD in anti-cervical cancer.Methods: CCK-8 assay was used for examining the proliferation of Hela cell incubated with SSD,the most popular cell line of cervical cancer.And then,the flow cytometry was used for performing cell cycleand apoptosis of Hela cell affected by SSD and,at last,the real time PCR was utilized for detecting the expressions of associated genes.Results: Compared to control,SSD could significantly inhibit the proliferation(P〈0.01) and induce the apoptosis(P〈0.05) and arrest the cell cycle at G1 phase in Hela cell.Next,we found that levels of cyclin D1 and Cyclin E,both cell cycle genes,were decreased.At the same time,apoptosis-associated genes Bax and Bcl-2 had opponent levels.Conclusion: SSD inhibits the proliferation of cervical cell Hela cell though blocking of cell cycle at G1 phase and induction of apoptosis.
