摘要
目的:观察银杏叶提取物(GBE)对急性心肌梗死(AMI)后转化生长因子-β1表达的影响。方法:将30只雄性SD大鼠随机分为假手术组,模型组,银杏叶组3组。按文献方法假手术组左冠状动脉前降支下只穿线不打结,另外两组均结扎左冠状动脉前降支。其中银杏叶组腹腔注银杏叶提取物2 m L/kg,假手术组和模型组分别腹腔注射等量生理盐水,连续注射4周后,HE染色观察心肌组织形态,免疫组织化学检测转化生长因子-β1(TGF-β1)、I型胶原蛋白的表达。结果:银杏叶治疗组心功能明显优于模型组,心脏重量/体重、左室重量/体重低于模型组(P<0.05),TGF-β1、I型胶原蛋白的表达明显低于模型组(P<0.01)。结论:银杏叶提取物能够通过降低TGF-β1、I型胶原蛋白的表达抑制急性心肌梗死引起的心室重构。
Objective: To observe the protective effects of the extract of Ginkgo biloba extract(GBE) on experimental cardiac remodeling in rats induced by acute cardiac infarction and explore the mechanism of anti-cardiac remodeling of GBE.Methods: Rats were randomly divided into three groups(n = 30):sham operation group(group A),acute cardiac infarction model group(group B) and acute cardiac infarction model + GBE(2 m L/kg) treatment group(group C).The acute cardiac infarction model was reproduced by ligaturing the left anterior descending artery excluding the sham operation group which did not ligation but only completed the operational process.Each group was further subdivided into treatment regimens lasting 4 weeks.The protein expressions of rat myocardial TGF-β1 and type I collagen were detected with immunohistochemical method.Results:The heart weight/body weight ratio and the left ventricle weight/body weight ratio were significantly decreased(P〈0.05).Compared with group B,the protein expression levels of TGF-β1 and type I collagen in group C were significantly decreased(P〈0.01).Conclusions: GBE could inhibit experimental rats cardiac remodeling after acute cardiac infarction via reducing the expressions of TGF-β1 and type I collagen in myocardial cells.
