粗壮女贞提取物对2型糖尿病小鼠的降血糖效果及作用机制 被引量:3

摘要 目的:研究粗壮女贞(Ligustrum robustum BL)对2型糖尿病小鼠的降血糖机制。方法:采用腹腔注射四氧嘧啶(200 mg·kg-1)建立小鼠2型糖尿病模型,实验分为正常对照组、阳性对照组、高、中、低剂量组(24.0g·kg-1·d-1、12.0 g·kg-1·d-1、8.0 g·kg-1·d-1),ELISA试剂盒检测2型糖尿病小鼠血清中血糖值水平(GLU)及血清胰岛素水平,Western blotting法检测小鼠脂肪组织中胰岛素受体(IR-β)、蛋白激酶B(PKB)和葡萄糖转运蛋白4(Glut4)蛋白表达。结果:粗壮女贞提取物能显著降低2型糖尿病小鼠血糖水平(GLU),增加血清胰岛素含量(P<0.05,P<0.01),促进脂肪中胰岛素受体(IR-β)、蛋白激酶B(PKB)和葡萄糖转运蛋白4(Glut4)蛋白的表达。结论:粗壮女贞提取物对2型糖尿病小鼠具有显著的降血糖作用,可显著改善2型糖尿病小鼠糖脂代谢并抑制胰岛素抵抗。 Objective: To study the effect of Ligustrum robustum(Ligustrum robustum BL) hypoglycemic mechanism on mice with type 2 diabetes mellitus.Methods: The intraperitoneal injection of four oxygen pyrimidine 200 mg·kg^-1to establish the mice model of type 2 diabetes.The rats were divided into normal control group,positive control group,high,medium and low dose groups(24.0 g·kg^-1· d^-1,12.0 g·kg^-1· d^-1,8.0 g·kg^-1· d-1).ELISA kit was used for the detection of type 2 diabetic mice serum glucose(Glu) levels and serum insulin level.Western blotting was used to detect the mice adipose tissue of insulin receptor(IR beta),protein kinase B(PKB) and glucose transport protein 4(GLUT4) protein expression.Results: Ligustrum robustum extract could significantly decrease the type 2 diabetic mice blood glucose levels(Glu),increase the serum insulin levels(P〈0.05,P〈0.01) and promote fat in insulin receptor(IR beta),protein kinase B(PKB) and glucose transport protein 4(GLUT4) protein expression.Conclusion: Ligustrum robustum extract on mice with type 2 diabetes has remarkable effect on reducing blood sugar,improving type 2 diabetes glucose and lipid metabolism in mice and inhibiting insulin resistance.
