摘要
目的:皮下接种H22肝癌细胞株构建小鼠H22肝癌移植瘤模型,研究兖州卷柏总生物碱对小鼠抗肿瘤效果。方法:将H22肝癌移植瘤小鼠随机分5组,Ⅰ组为NS阴性对照组,每天一次性灌胃等量生理盐水;Ⅱ组按20 mg/kg剂量每天一次性灌胃环磷酰胺(CTX)水溶液;Ⅲ、Ⅳ和Ⅴ组分别按40、80、120 mg/kg剂量每天一次性灌胃兖州卷柏总生物碱溶液。连续2周,末次给药24 h后,称体重,观察治疗前后小鼠体重变化;颈椎脱臼处死小鼠,剖取腋下瘤组织、肝肝、脾脏和胸腺组织,分别称重,计算抑瘤率和胸脾指数。取部分肿瘤组织10%甲醛固定,切片,HE染色,观察肿瘤组织病理学变化。结果:各剂量的兖州卷柏生物碱均能延长H22肝癌移植瘤小鼠生存时间和抑制肿瘤生长,且各剂量组中抑瘤率呈现剂量-效应关系。结论:兖州卷柏生物碱对小鼠H22移植瘤具有抑制效果。
Objective: To investigate the antitumor effect of total alkaloids from Selaginella involvens on transplantation tumor of hepatoma H22 in mice and the transplantation tumor of hepatoma H22 in mice was established.Methods:The animals were randomly divided into five groups and treated as follows:groupⅠwas negative control group and was given commensurable normal saline.GroupⅡwas positive control group and was were given 50 mg/kg cyclophosphamide(CTX).Group Ⅲ,Ⅳ and Ⅴ were given total alkaloids from S.involvens with the dose of 40 mg/kg,80 mg/kg,120 mg/kg respectively.All mice were treated for 2 weeks.24 h after the last administration,all animals were weighted and tumors,livers,thymus and spleens were measured and the inhibitory rate and the index of thymus and spleen were analyzed.For histological analysis,the tumors were fixed in 10% formaldehyde at room temperature.And then 5 μm thick serial sections were cut using microtome and stained with haematoxylin-eosin.Results: Administration of total alkaloids from S.involvens significantly prolonged the survival time and inhibited the growth of transplantation tumor of hepatoma H22 in mice and the inhibitory rate showed a dose-response relationship.Conclusions: Total alkaloid from Selaginella involvens could inhibit the growth of transplantation tumor of hepatoma H22 in mice.
