丹参对鱼胆毒素致急性肾损伤保护作用的体内外实验研究 被引量:4

Experimental Study on Protective Effect of Salvia Miltiorrhiza on Acute Renal Injury Induced by Fish Bile Poison in Vivo and in Vitro

摘要 目的:观察丹参注射液对草鱼胆汁致急性肾损伤的影响。方法:健康雄性SD大鼠,随机平分为对照组、模型组、丹参干预组。采用草鱼胆汁灌胃造模,丹参腹腔给药干预。连续不同时点检测血肾功,尿测肾损伤分子-1(KIM-1)、NAG,同时常规肾脏病理检查及免疫组化检测KIM-1表达。从新鲜草鱼胆汁中提取鲤醇硫酸酯钠,通过体外培养人肾小管上皮细胞(HK-2),应用MTT法检测不同浓度丹参干预染毒HK-2细胞的活力。结果:模型组大鼠血肌酐,尿KIM-1及NAG均于鱼胆中毒6 h后明显升高,血尿素于中毒12 h后明显升高,丹参干预组24 h后上述指标明显下降(P<0.05);模型组大鼠肾小管上皮细胞变性、坏死、脱落,中毒6 h后肾组织近曲小管KIM-1表达增强(P<0.05),丹参干预组大鼠肾小管病变明显减轻,中毒24 h后肾小管KIM-1表达明显下调(P<0.05)。鲤醇硫酸酯钠染毒HK-2细胞存活率明显低于对照组(P<0.05),丹参干预鲤醇硫酸酯钠染毒HK-2细胞存活率明显增加(P<0.05),且呈现剂量依赖关系。结论:丹参注射液对草鱼胆汁致急性肾小管损伤有一定保护作用。 Objective:To investigate the effect of Salvia miltiorrhiza injection on acute renal injury induced by grass carp bile.Methods: The male SD rats were randomly divided into control group,model group and Salvia miltiorrhiza group.Grass carp bile was gavaged to the rats and the Salvia miltiorrhiza injection was injected intraperitoneally.Serum renal function,urine KIM-1 and NAG were determined.Routine renal pathology was completed and renal KIM-1 expression was detected by immunohistochemistry.Sodium Cyprinol Sulphate was extracted from the fresh grass carp bile,human renal tubular epithelial cells(HK-2) was cultured in vitro and the activity of HK-2 cells was detected by MTT assay.Results:Compared with the model group,the Scr,urine KIM-1 and NAG were increased significantly after 6hours.The denatured,necrotic and deciduous renal tubular epithelial cells were observed and expression of KIM-1 in renal tissue was increased after 6 hours of poisoning in the model group(P〈0.05).Compared with the model group,the renal tubular lesions were significantly decreased and the expression of KIM-1 in renal tubular was significantly decreased after 24 h of poisoning in rats treated with Salvia miltiorrhiza(P〈0.05).The survival rate of HK-2 cells in the model group was significantly lower than that in the other groups(P〈0.05).Compared with the model group,the survival rate of HK-2 cells was significantly increased in Salvia miltiorrhiza group(P〈0.05) and the dose-dependent relationship was presented.Conclusion:Salvia miltiorrhiza injection has a protective effect on acute renal tubular injury induced by grass carp bile.
