摘要
目的探讨前列腺素E2(PGE2)在前列腺间质活化过程中的作用。方法体外培养人前列腺间质细胞人正常前列腺基质永生化细胞(WPMY-1),用二甲基亚砜(DMSO)和10-9 mol/L前列腺素E2(PGE2)处理后进行免疫荧光染色,检测肌成纤维细胞表型的变化。实时定量聚合酶链反应(PCR)检测细胞中炎性因子白细胞介素-8(IL-8)的表达,并测定IL-8的浓度。结果 PGE2显著增加WPMY-1细胞中α平滑肌肌动蛋白(α-SMA)和波形蛋白共定位的细胞比例。PGE2促进WPMY-1细胞中IL-8的表达。结论 PGE2能增加体外培养的WPMY-1肌成纤维细胞表型,促进IL-8的表达,在前列腺癌发生、发展中具有重要的作用。
Objective To investigate the effects of prostaglandin E2(PGE2)in prostate mesenchymal cell activation.MethodsTo culture human prostate stromal cells WPMY-1in vitro,implement immunofluorescence staining after treating with DMSO and10-9 mol/L PGE2 and detect the change of the myofibroblast phenotype.The expression of cellular inflammatory factor interleukin8(IL-8)was detected by real-time quantitative PCR and the concentration of IL-8was detected.Results PGE2 significantly increased the cellular proportion of colocalization with alpha SMA and Vimentin in WPMY-1cells.PGE2 promoted the expression of IL-8in WPMY-1cells.Conclusion PGE2 can increase myofibroblast phenotype in human prostate mesenchymal cells WPMY-1in vitro culture,promotes the IL-8expression,which has an important role in the occurrence and development of prostate cancer.
