摘要
目的观察山香圆总黄酮对LPS诱导的RAW264.7细胞中iNOS、COX-2及NF-кB表达的抑制作用。方法采用Griess法检测山香圆总黄酮对NO生成的影响,通过westernblot分析i NOS、COX-2及NF-кB的表达。结果山香圆总黄酮能显著抑制RAW264.7细胞中NO的生成以及iNOS、COX-2及NF-кB蛋白的表达,且随着浓度的增加,抑制效果明显。结论山香圆总黄酮的抗炎作用是通过下调NF-кB的活性进而抑制i NOS、COX-2的表达来实现的。
Objective To explore the inhibitory effect of total flavonoids of Turpinia arguta Seem(TFS) on inducible nitric oxide synthase(iNOS) ,cyclooxygenase(COX) -2 ,nuclear factor (NF) -κB protein expression in lipopolysaccharides(LPS) -stimulated RAW264.7 ceils. Methods the effect of TFS on the product of NO in RAW264.7 cells was measured by the Griess reagent ; and the expression of iNOS , COX - 2 , NF -κB protein in RAW264.7 cells were analyzed by Western - blot method. Results TFS inhibited the LPS - stimulated NO synthesis in RAW 264.7 cells. The expression of iNOS, COX -2, NF - xB pro- tein were also decreased by TFS in the RAW264.7 cell, and the inhibitory effect was enhanced with the increasing of TFS concentration. Conclusion The results show that the anti - inflammatory effects of TFS are due to the down - regulation of NF - κB and the Consequen expressional suppressions of iNOS and COX -2.
