响应曲面法优化纳滤浓缩延胡索水提液的工艺研究 被引量:2

摘要 目的利用响应面分析方法优化延胡索提取液的纳滤浓缩工艺。方法以延胡索乙素截留率为指标,在明确压力、温度、平衡体积等因素影响的基础上,采用Box-Behnken中心组和设计建立数学模型,考察截留分子量、浓度、p H对延胡索乙素截留率的影响。结果优选出最佳纳滤浓缩条件为:纳滤膜截留分子量450 Da,药液浓度192.68μg/m L,p H 4.72,延胡索乙素理论截留率为95.00%,实测截留率为(93.99±1.29)%。结论该模型可以较好的预测延胡索乙素的截留率,响应面法对中药纳滤浓缩的参数优选合理可行,为热敏性中药成分的纳滤浓缩提供基础。 Objective Response surface analysis methodology was used to optimize the concentrate process of COrydalis yanhusuo extract by nanofihration technique. Methods On the basis of the influences of pressure, temperature and circulating volume, the retention rate of tetrahydropaimatine was selected as index, molecular weight cut - off of nanofiltration membrane, concentration and pH were selected as influencing factors during concentrate process. The experiment mathematical model was arranged according to Box - Behnken central composite experiment design. Results The optimal concentrate conditions were as following: molecular weight cut off450 Da, pH 4.72, concentration 192.68 μg/mL, the predicted value of retention rate of tetrahydropalmatine was 95.00% under the optimum conditions, and the experimental value was (93.99 ± 1.29) %. Conclusion The combination of Box - Behnken design and response surface analysis can well optimize the concentrate process of tetrahydropalmatine by nanofiltra- tion, and the results provide the basis for nanofihration concentrate for heat - sensitive traditional Chinese medicine.
