摘要
目的对土壤放线菌SSPRC-1次级代谢产物中杀虫和抑菌活性物质进行研究。方法对发酵液通过正丁醇萃取、硅胶柱色谱、HPLC等手段进行分离纯化,利用质谱、核磁共振谱等现代波谱分析方法鉴定活性化合物结构。结果从放线菌SSPRC-1发酵液中分离得到3个活性化合物SSPRC-1-1、SSPRC-1-2和SSPRC-1-3,确定SSPRC-1-1分子式为C15H10O4,是异黄酮类物质,在天然产物中未曾发现,仅有文献报道可通过化学合成得到,暂命名为isoflavones H;SSPRC-1-2分子式为C15H10O5,与异黄酮类物质三羟基异黄酮genistein相同;SSPRC-1-3分子式为C25H37NO4,与杀粉蝶毒素A结构相同。结论化合物SSPRC-1-1和SSPRC-1-2对Pyricularia oryzae的MIC分别为125和24μg/m L,化合物SSPRC-1-3对黏虫具有较好的毒杀作用。
Objective To investigate the activities of the secondary metabolites against Pyricularia oryzae and Mythimna separate (Walker) from the strain Streptomyces SSPRC-1. Methods By bioassay-guided isolation and purification, the compounds were isolated and purified from fermentation with n-butyl, silica gel column chromatography and HPLC, and their structures were elucidated with the help of the spectra data analysis of UV, ESI- MS and NMR. Results Three compounds were isolated from the fermentation broth of Streptomyces SSPRC-1 and named as SSPRC-1-1, SSPRC-1-2 and SSPRC-1-3. The molecular formula of the compound SSPRC-1-1 was confirmed as C15H1004, which was identified as a new natural compound named isoflavone H. The molecular formulas of the compound SSPRC-1-2 and SSPRC-1-3 were confirmed as C15H1005 and C25H37NO4, the same as the known compound genistein and piericidin, respectively. Conclusion The MIC of two compounds SSPRC-1-1 and SSPRC-1-2 against P. oryzae were 125 and 24μg/mL. Compound SSPRC-1-3 showed good bioactive against Mythimna separate.
