This paper reviewed the methods of synthesizing tenofovir alafenamide, and analyzed different synthetic routes of key intermediates. The advantages and disadvantages of each route were then evaluated according to yields, costs and process difficulties. The synthetic route with large industrial production value was selected to provide references and methods for the further research oftenofovir alafenamide.
