ADAM33基因T1位点多态性与内蒙古地区蒙、汉民族人群哮喘的相关性研究 被引量:2

摘要 目的探讨解整合素-金属蛋白酶33(ADAM33)基因T1位点(rs2280091)不同基因型及等位基因的分布频率与内蒙古地区蒙、汉民族支气管哮喘的关系。方法选用限制性片段长度多态性(PCRRFLP)方法对汉族哮喘患者112例、蒙古族哮喘患者105例进行ADAM33基因多态性的检测,并分别与110例健康汉族和108例健康蒙族进行比较,筛选有意义基因。结果内蒙古地区蒙、汉族人群均可检出T1位点3种基因型(AA、AG、GG),在蒙、汉族支气管哮喘组分布频率分别与健康对照组比较差异有统计学意义(P<0.05),汉族哮喘组AG、GG基因型OR值分别为0.183、0.38,蒙古族哮喘组AG、GG基因型OR值分别为0.295、0.851;蒙、汉族支气管哮喘组T1位点等位基因A和G基因频率分别与健康对照组比较差异有统计学意义(P<0.05),G等位基因OR值及95%可信区间分别为0.372(0.190-0.729)、0.237(0.111-0.509)。结论内蒙古地区ADAM33基因T1位点基因多态性与内蒙古地区蒙、汉族人群哮喘发病可能相关。 Objective To explore the association of the polymorphism of TI locus allele (rs2280091) in recombinant A disintegrin and metalloprotease 33 ADAM33 gene with bronchial asthma in Inner Mongolia Han and Mongolian population. Methods 112 screened Han population patients with asthma (the asthma group A) and 110 unrelated healthy Han population (the healthy control group A) were selected. 105 screened Mongolian population patients with asthma ( the asthma group B ) and 108 unrelated healthy Mongolian population ( the healthy control group B) were selected. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to determine polymorphism of T1 locus allele in ADAM33 gene in all groups. Results Three genotypes (AA, AG and GG) were found in Inner Mongolia Han and Mongolian population. The differences of distribution frequency in the Mongolia, Han bronchial asthma groups were significant while compared with the control group ( P 〈0. 05). The OR value of AG and GG genotype was 0. 183 and 0. 38 in the asthma group A, and 0. 295 and 0. 851 in the asthma group B. There were significant differences in T1 gene frequency loci alleles A and G between the bronchial asthma groups and the control group (P 〈 0.05 ). The OR value and the 95% confidence of G allele interval was 0. 372 (0. 190 -0. 729) in the asthma group B, and 0. 237 (0. 111-0. 509) in the asthma group A. Conclusion T1 AD- AM33 gene locus gene polymorphism is associated with asthma in Inner Mongolia Han and Mongolian population.
