摘要
目的研究甜菜碱对异丙肾上腺素诱导大鼠急性缺血性心肌损伤炎症因子表达的抑制作用。方法用甜菜碱(100、200、400 mg·kg-1·d-1)灌胃大鼠40 d,第39、40天于皮下注射85 mg·kg-1·d-1异丙肾上腺素;取大鼠左心室的心肌组织,采用蛋白芯片技术再结合酶联免疫检测方法,研究甜菜碱对急性缺血心肌炎症因子表达的影响。结果甜菜碱对心肌急性缺血性损伤具有保护作用。与正常组比较,试验组中急性缺血心肌炎症因子β-NGF、TIMP-1、ICAM-1、MCP-1的表达增加,IL-1β、IL-4、VEGF的表达减少;给予大鼠甜菜碱(200、400 mg·kg-1·d-1),对缺血心肌MCP-1和ICAM-1的高表达具有抑制作用。结论甜菜碱可通过抑制心肌MCP-1和ICAM-1的表达对急性心肌缺血性损伤具有保护作用。
OBJECTIVE To investigate the inhibitory effect of betaine on expression of inflammation factor in isoproterenol - induced acute myocardial ischemia in rats. METHODS Betaine (100,200,400 mg·kg^-1·d^-1 ) was administered orally once a day for 40 days. Acute myocardial injury was induced by subcutaneous injection of isoproterenol (Isn) (85 mg·kg^-1·d^-1) in rats on the 39 th and 40 th day. Expression of the inflammation factor in left ventricle was measured using of protein - chip technique and enzyme - linked immunosorbent assay. RESULTS Betaine exerted cardioprotective effects on Iso - induced myocardial ischemia in rats. Compared to the control group,the contents of β- NGF,TIMP - 1, ICAM - 1 and MCP - 1 were increased, and the levels of IL - 1β, IL -4 and VEGF were decreased in Iso treated rats. Oral administration of betaine( 200,400 mg·kg^-1·d^-1 ) significantly decreased the high level of MCP - 1 and ICAM - 1. CONCLUSION Betaine can protect myocardium against isoproterenol - induced ischemia through the inhibition of MCP - 1 and ICAM - 1 level.
