摘要
目的分离合欢皮中的皂苷物质,并评价其对人脐静脉内皮细胞(EA.hy926)增殖的影响。方法以细胞活性为指标,依次采用D101大孔树脂、硅胶柱、反相C18色谱柱从合欢皮正丁醇相中分离得到活性组分H5;采用UPLC-TOF-MS分析,确定此组分中所含的主要成分。结果组分H5中主要皂苷类物质为julibroside J5、julibroside J8、isomer of J5和isomer of J8,总含量为79.15%,且对EA.hy926细胞的增殖具有抑制作用(IC50=7.82±1.59μg·m L-1)。结论成功分离得到了具有抗血管新生作用的合欢皮皂苷物质。
OBJECTIVE To separate and purify saponins from Albizzia julibrissin and to investigate their effect to human umbilical vascular endothelial cells (EA. hy926). METHODS The cell viability was used meansured. An anti - angiogenesis component H5, based on cell viability,was obtained from n -butanol fraction of ethanol extracts of A. julibrissin using D101 macroporous resin elution, silica gel column chromatography and RP - 18 column chromatography. Then, the main compounds in H5 were determined using UPLC - TOF - MS technique. RESULTS The main compounds in H5 were julibroside J5 ,julibroside Js,isomer of J5 and isomer of Js. The content of all saponins was 79.15% measured using HPLC analysis. Compounds Hs exhibited an inhibition activity to EA. hy926 cells proliferation of IC50 with the value of 7.82 ±1.59 μg·m L^-1. CONCLUSION The saponins from A. julibrissin with the anti - angiogenesis activity has been separated successfully.
