摘要
目的采用HPLC法测定清热解毒口服液中黄芩苷、绿原酸、连翘苷、连翘酯苷A的含量。方法采用Zorbax Eclipse XDB C18色谱柱(150 mm×4.6 mm,5μm),柱温30℃,流动相为乙腈-0.3%磷酸溶液,梯度洗脱,流速1.0 m L·min^(-1),检测波长324 nm(绿原酸、连翘酯苷A)、276 nm(黄芩苷、连翘苷)。结果黄芩苷、绿原酸、连翘苷、连翘酯苷A的线性范围分别为0.182~0.912、0.061~0.304、0.037~0.183、0.013~0.065μg,加样回收率分别为98.82%、96.94%、105.6%、95.74%(n=6)。结论所用方法操作简单,为全面控制清热解毒口服液的质量提供了一种可靠的检测方法。
OBJECTIVE To determine baicalin, chlorogenic acid and forsythin and forsythoside A in Qingre Jiedu oral liquid(QJOL) by HPLC. METHODS The Agilent Zorbax Eclipse XDB C18 column ( 150 mm × 4.6 mm,5 μm) was used with acetonitrile - 0.3% phosphoric acid solution by gradient elution. The flow rate was 1.0 mL· min^-1 with the column temperature of 30 ℃. The detection wavelength was 324 nm for chlorogenic acid, forsythoside A and 276 nm for baicalin and forsythin. RESULTS The linear range of baicalin,chlorogenic acid,forsythin and forsythoside A was in the ranges of 0. 182 -0. 912,0. 061 -0. 304,0. 037 -0. 183 and 0.013 -0.065 μg. The average recoveries of baicalin, chlorogenic acid ,forsythin and forsythoside A were 98.82% ,96.94%, 105.6% and 95.74% (n = 6). CONCLUSION The method has the advantages of simple operation, comprehensive and effective quality control of QJOL. And the method provides a reliable detection method.
