冬虫夏草菌的分离、鉴定及菌丝体多糖含量测定 被引量:2

Isolation and identification of Cordyceps strains from Naqu Cordyceps sinensis and determination of polysaccharides

摘要 对存在于那曲冬虫夏草中的冬虫夏草菌进行分离鉴定,并测定其菌丝体多糖含量。于新鲜那曲冬虫夏草子座中初步分离得到冬虫夏草菌,用梯度划线法进一步纯化,并进行菌落形态观察及ITS序列分析,经液体发酵后,采用苯酚-硫酸法测定菌丝体多糖含量。结合菌株的形态和ITS序列鉴定结果,将菌株CC-109鉴定为Verticillium sp.,经测定菌丝体多糖含量为3.742%。 The aim of this experiment was to isolate and identify Cordyceps strains from Naqu Cordyceps sinensis, determine the content of polysaccharides. Cordyceps strains were separated preliminarily from storma of fresh Naqu Cordyceps sinensis and purified by gradient streaking. The colony morphology was observed and ITS sequence was analyzed. Phenol-sulfuric acid method was used to determine the content of polysaccharides. CC-109 strain was identificated as Verticillium sp according to morphology of strain and ITS sequence. The polysaccharide contents of strain CC-109 was 3.742%.
