摘要
目的:观察独活寄生汤对胶原性关节炎(CIA)大鼠膝关节软骨细胞的影响。方法:用牛Ⅱ型胶原诱导大鼠关节炎模型,分为模型组、独活寄生汤组和布洛芬组3组,另设正常组。分别予独活寄生汤、布洛芬及生理盐水灌胃,持续30 d后取材。关节软骨组织切片采用番红-快绿染色,进行关节软骨组织学评分;透射电镜观察软骨细胞微结构变化。结果:独活寄生汤组软骨组织学评分提示软骨退变减轻,与模型组、布洛芬组比较,差异有统计学意义(P<0.05)。独活寄生汤组透射电镜下软骨细胞线粒体和高尔基体数目均较模型组增加,差异有统计学意义(P<0.05)。结论:独活寄生汤可缓解胶原性关节炎大鼠软骨细胞退变。
Objective: To observe the effect of Duhuo Jisheng Decoction on knee articular chondrocytes in collageninduced arthritis( CIA) rats. Methods: Type Ⅱ collagen was used to induce CIA model in rats. Model rats were divided into three groups: model group,Duhuo Jisheng Decoction group and ibuprofen group,normal group was set alone. Corresponding drugs were given orally to the Duhuo Jisheng Decoction group and ibuprofen group,and normal saline were given orally to the other two groups. After 30 days the samples were collected. The sections of articular cartilage tissue were stained with safraninorange / light-green staining and the histological score of articular cartilage was studied; the microstructure changes of chondrocytes were observed by transmission electron microscope. Results: The results of histological score of articular cartilage suggested that chondrocyte degradation was relieved after the treatment with Duhuo Jisheng Decoction,with statistically significant differences compared with the model group and ibuprofen group( P〈0. 05). Compared with the model group,the numbers of mitochondria and Golgi apparatus of chondrocytes in the Duhuo Jisheng Decoction group were increased,with statistically significant differences( P〈0. 05). Conclusion: Duhuo Jisheng Decoction may relieve the chondrocyte degradation in collagen-induced arthritis rats.
