摘要
目的:观察淫羊藿苷(ICA)调节ApoE基因敲除小鼠脂质代谢的作用,从而探讨其改善动脉粥样硬化病理环境的疗效机制。方法:8周龄ApoE基因敲除小鼠22只随机分为3组。其中,空白组7只,予普通饮食及0.9%NaCl溶液腹腔注射;对照组7只,予高脂饮食及0.9%NaCl溶液腹腔注射;实验组8只,予高脂饮食及ICA每日200 mg/kg腹腔注射。各干预8周后观察各组小鼠体质量、白色脂肪质量、白色脂肪/体质量比例、血清胆固醇(TC)、三酰甘油(TG)、血糖水平,用实时荧光定量PCR(RT-q PCR)方法检测白色脂肪脂蛋白酯酶(LPL)mRNA表达。结果:与空白组比较,对照组小鼠体质量、白色脂肪质量、白色脂肪/体质重比例明显增高(P<0.05);与对照组比较,实验组小鼠体质量、白色脂肪质量、白色脂肪/体质重比例均有所降低(P<0.05)。与空白组比较,对照组TC、TG水平升高(P<0.05);与对照组比较,实验组血糖、TC、TG水平均明显降低(P<0.05)。与空白组比较,对照组小鼠LPL mRNA表达明显降低(P<0.05);与对照组比较,实验组小鼠LPL mRNA表达明显升高(P<0.05)。结论:ICA可能通过降低ApoE基因敲除小鼠血脂、血糖水平,同时增加白色脂肪LPL表达增加脂肪分解代谢,从而减少白色脂肪堆积,改善其动脉粥样硬化病理环境。
Objective: To observe the effects of Icariin( ICA) on white adipose tissue metabolism in ApoE knocked mice and discuss its mechanism on improving the pathological environment of atherosclerosis. Methods: 22 ApoE knocked mice that8 weeks old were randomly divided into three groups: 7 mice in the blank group were administrated with normal diet and 0. 9%NaCl for intraperitoneal injection; 7 mice in the control group were administrated with high fat diet and 0. 9% NaCl for intraperitoneal injection; 8 mice in the experimental group were administrated with high fat diet and ICA for intraperitoneal injection at the dose of 200 mg / kg daily. After 8 weeks,the body weight,white fat weight,ratio of white fat and body,serum level of cholesterol( TC),triacylglycerol( TG) and blood glucose of mice in all groups were observed,and the mRNA expression of lipoprotein lipase( LPL) was determined by real time fluorescence quantitative PCR( RT-q PCR). Results: Compared with the blank group,the body weight,white fat weight and the ratio of white fat and body in the control group were obviously increased( P〈0. 05); compared with the control group,the body weight,white fat weight and the ratio of white fat and body in the experimental group were all decreased( P〈0. 05). Compared with the blank group,the levels of TC and TG in the control group were increased( P〈0. 05); compared with the control group,the levels of blood glucose,TC and TG in the experimental group were all obviously decreased( P〈0. 05). Compared with the blank group,the mRNA expression of LPL in the control group were obviously decreased( P〈0. 05); compared with the control group,the mRNA expression of LPL in the experimental group were obviously increased( P〈0. 05). Conclusion: ICA may reduce the serum levels of TC,TG and glucose in ApoE knocked mice and increase LPL expression of white adipose tissue to promote the lipoclasis,which consequently alleviate the accumulation of white fat,improve the pathological environment of atherosclerosis.
