摘要
对不同产地、不同采收期及不同部位叶下珠药材没食子酸、咖啡酸含量进行测定并比较分析。分析采用Hypersil ODS2(250mm×4.60mm,5um)色谱柱,以乙腈(A)-0.1%磷酸水溶液(B)二元系统梯度洗脱,柱温25℃,流量0.8mL/min,检测波长280nm。没食子酸在0.157~1.570mg/g(r=0.998 7)呈良好的线性关系,加样回收率(n=5)为98.7%;咖啡酸在0.283~2.830mg/g(r=0.998 3)呈良好的线性关系,加样回收率(n=5)为99.4%。结果表明:不同产地叶下珠药材没食子酸、咖啡酸的含量有较大差异,没食子酸的含量在0.533~1.039mg/g,咖啡酸的含量在0.880~3.297mg/g;不同采收期叶下珠药材没食子酸、咖啡酸含量在10月5日最高,建议叶下珠药材采收期为每年的10月上旬;不同部位叶下珠药材没食子酸、咖啡酸含量差异较大,叶含量最高,根最低,建议叶下珠药材只采收地上部分。本试验建立HPLC法同时测定叶下珠药材中没食子酸、咖啡酸的含量,该方法简便、准确、重现性好,可为叶下珠药材质量评价及质量控制提供依据。
Determination and comparative analysis of the contents of gallic acid and caffeic acid in Phyllanthus urinaria L.from different habitats,of different harvest time,different parts was done by HPLC.The analysis was carried out on a Hypersil ODS2column(250mm×4.60 mm,5μm),mobile phase was acetonitrile(A)-0.1% phosphoric acid aqueous(B)with gradient elution,column temperature was maintained at 25℃,flow rate was 0.8ml/min and detection wavelength was set at 280 nm.Gallic acid showed good linear relationships within the ranges of 0.157~1.570mg/g(r=0.998 7),whose average recoveries(n=5)were 98.7%.Caffeic acid showed good linear relationships within the ranges of 0.283~2.830mg/g(r=0.998 3),whose average recoveries(n=5)were 99.4%.The results were as follows:the content of gallic acid and caffeic acid in Phyllanthus urinaria L.varied greatly with different habitats.The content of gallic acid was from 0.533mg/g to 1.039mg/g,the content of caffeic acid was from 0.880mg/g to 3.297mg/g;the content of gallic acid and caffeic acid reached the highest in October 5th,it was suggested the best harvest time is early October each year;the contents of gallic acid and caffeic acid were obviously different in Phyllanthus urinaria L.from different parts,the highest was from leaf,the contents of gallic acid and caffeic from root was very low,therefore,suggested only collecting the aerial parts.This method is simple,accurate with good reproducibility,which can be used for the quality control of Phyllanthus urinaria L.
