ZD制剂对小鼠巨噬细胞促炎性介质的影响 被引量:4

THE IMPACT OF ZD PREPARATION ON PRO-INFLAMMATORY FACTORS IN MICE MACROPHAGES

摘要 目的:研究ZD制剂在体外的抗炎机制。方法:通过脂多糖(LPS)刺激RAW264.7细胞系建立炎症细胞模型,使用5种不同浓度的ZD制剂进行试验,分别采用Griess反应测定细胞上清液中NO水平,ELISA法检测ZD制剂干预前后细胞上清液中TNF-α、IL-1β、IL-6的浓度。结果:用ZD制剂干预后,上清液中NO、TNF-α、IL-1β、IL-6含量与模型组相比均有下降,并存在剂量依赖关系。结论:因此,中药复方汤剂ZD制剂的体外抗炎作用机理之一是抑制NO、TNF-α、IL-1β、IL-6等炎性介质的产生。 [Objective] To investigate the anti - inflammatory mechanism of ZD preparation in vitro. [ Method] The cell model was established by lipopolysaccharide(LPS) induction on the RAW264.7 macrophages cell line. Using five kinds of different concentrations of ZD preparation experiment. Griess assay was used to measure the secretion of NO. ELISA assay was used to measure the secretion of TNF - α, IL - 1β and IL - 6 in supernatant of ZD preparation treated and untreated maerophages. [ Results ] The secretion levels of NO, TNF - α, IL - 1β and IL - 6 were significantly lower in the supernatant of ZD preparation treated macrophages than those in controls. [Conclusion] ZD preparation could inhibit inflammation by down- regulating the secretion of pro -inflammatory factors.
